Objective:The clinical efficacy of Jianpi Xiaoke Decoction in the treatment of patients with spleen deficiency and blood stasis-heat type of Xiaoke was retrospectively analyzed, and its molecular mechanism was discussed based on in vitro experiments and Shengxin analysis.Methods:1.Retrospective study: The medical records of patients with spleen deficiency and blood stasis-heat type of diabetes mellitus who met the inclusion criteria were collected through the medical record management system of the famous old Chinese medicine network in Shandong Province, and the patients were grouped according to the grouping criteria. The relevant information of patients was recorded, and the efficacy and adverse reactions of different groups were statistically analyzed.SPSS26.0 was used for statistical analysis of the data.2.Experimental study: Min6 islet cell lines were intervened and modeled with different concentrations of complete medium according to the normal group, model group and traditional Chinese medicine group. The apoptosis of islet βcells was observed in vitro, and the molecular mechanism of Jianpi Xiaoke Decoction was explored by Shengxin analysis.Results:1.Through retrospective analysis found that Chinese medicine group and control group after treatment in patients with blood lipids, blood glucose, insulin resistance, and TCM syndrome curative effect comparison differences were statistically significant(P < 0.05).2.Experimental study found that Jianpi Xiaoke Decoction can effectively improve the apoptosis of islet β cells, which may play a role through AGE-RAGE,PI3K-Akt,IL-17,HIF-1 and other related molecular pathways. Conclusion:The curative effect of Jianpi Xiaoke Decoction combined with metformin in the treatment of diabetes patients with spleen deficiency and blood stasis heat type is better than that of metformin alone. The combination therapy can significantly improve the blood lipid, blood glucose and insulin resistance of patients, and to some extent improve fatigue, abdominal distention and other symptoms, with good safety.Jianpi Xiaoke Decoction mainly improves islet β cell apoptosis by regulating AGE-RAGE signaling pathway, thereby delaying the progress of diabetes.
