| Objective: To explore the relationship between intestinal flora changes and serum IL6 concentration in acute asthma mouse model adds a new drug of choice for childhood asthma treatment.Method: 16 clean-grade healthy female BALB/c mice(20g±)aged 4-5 weeks were selected,and 8 mice were randomly selected as the normal control group and given intraperitoneal injection of 1ml of normal saline,and the remaining 8 mice were given intraperitoneal injection 200 ul of chicken ovalbumin solution(containing 10 ug of OVA,1mg of aluminum hydroxide gel,dissolved in 1ml of normal saline)was injected intraperitoneally on the 1st day,the 7th day and the 14 th day respectively to obtain sensitized mice.On the 21 st,22nd,and 23 rd days of the experiment,the mice were placed in the mouse nebulizer,and the normal saline containing 1% OVA was used to generate aerosol particles(about 3-5um in diameter)using an ultrasonic nebulizer box for 30 times each time.Minutes,once a day,let the mice inhale,and the asthma symptoms such as restlessness,cyanosis,cough,shortness of breath and other symptoms appear after the challenge,indicating that the asthma model was successfully prepared.The normal control group was inhaled by aerosol inhalation of normal saline under the same conditions.1ml of mouse eye blood was collected within24 hours after the experiment,and the serum IL-6 concentration was determined by ELISA.The specific primers for the bacterial r RNA V4 region(515F and 806R)were extracted by using the rapid DNA extraction method to extract mouse fecal DNA.The16 S r DNA gene was amplified by PCR,and the PCR products were mixed and purified before library preparation,and the sequencing results were subjected to bioinformatics analysis.The composition and bacterial abundance of intestinal flora in the two groups were observed.Univariate statistical analysis and Pearson correlation analysis were performed on serum IL6 levels and Lactobacillus abundance using SPSS version 22.0software.Results:1.Compared with the control group,the serum interleukin(IL-6)concentration of mice in the experimental group was significantly increased(control group: 3.45±1.04,experimental group: 15.72±3.03,P<0.001),the difference was statistically significant significance.2.The results of species difference analysis showed that there was little difference in the composition of intestinal flora in the two groups of samples.3.The results of Anosim suggest that the grouping of the gut microbiota structure(combined with the abundance information of ASVs in the samples)has a statistically significant difference under the weighted condition(R=0.34,P=0.0199).4.The results of Lef Se showed that the main bacterial group with specificity in the two groups was Lactbacillaceae.Compared with the control group,the Lactobacillus family was significantly reduced in the intestinal flora of the mice in the experimental group.5.The results of Pearson test showed that the serum IL-6 concentration of mice in the experimental group was negatively correlated with the abundance of Lactobacillus family(r=-0.883,P=0.035<0.05).Conclusion:The concentration of IL-6 in the serum of the asthma model mice was significantly increased,and the Lactbacillaceae(Lactbacillaceae)in the community composition of the intestinal flora was significantly reduced,and the two were negatively correlated,indicating that the Lactbacillaceae in the intestinal flora has an effect on asthma inflammation.the occurrence of certain interventions. |
