Association Of PNPLA3 Gene Polymorphisms With East Asain Population NAFLD Susceptibility:A Meta-Analysis Of Case-Control Study

Objective:To analyze the the association between PNPLA3 gene rs738409 polymorphism and susceptibility of NAFLD in esat-asain population.Methods: Databases including CKNI、Wanfang、baidu、Dbpia、NCBI、Embase were searched to identify the literatures concerning the association between PNPLA3 gene rs738409 polymorphism and susceptibility of NAFLD in east-asain population from the inception to January 1,2022.Meta-analysis was performed using Stata SE 12.0 software after quality assessment.Results:(1)A total of 35 case-control studies from 33 literatures were included,involving 9900NAFLD cases and 13780 controls.(2)Meta-analysis shows that except additive model,allele model,dominant gene model,recessive model and codominant gene model all showed that the PNPLA3 gene rs2976392polymorphism was associated with an increased risk of NAFLD in east-asain population.In additive model rs738409 polymorphism may reduce risk of NAFLD(OR=0.951(0.901,1.004)P= 0.071).(3)Age subgroup : In adult group,except additive model,all gene type model showed that the PNPLA3 gene rs2976392 polymorphism was associated with an increased risk of NAFLD in adult group.but in additive model,PNPLA3 rs738409 polymorphism could reduce the risk of NAFLD(OR=0.938(0.885,0.993)P=0.029).In child group,except in heterozygote model may increase the risk of NAFLD(OR=1.276(0.853,1.909)P=0.235),rset genetype model showes that the PNPLA3 gene rs2976392 polymorphism was associated with an increased risk of NAFLD in child group.(4)Nation Subtype:In chinese and korea group,except additive model,all gene type model showed that the PNPLA3 gene rs2976392 polymorphism was associated with an increased risk of NAFLD,but in additive model,rs738409 polymorphism could reduce risk of NAFLD in chinese population(OR=0.936(0.880,0.995)P=0.034).In additive model,the PNPLA3 gene rs2976392 polymorphism may associated with an decreased risk of NAFLD in japan group,but there dont have statistically significant.(5)Chinese Subtype: Except additive model,all gene type model showed that the PNPLA3gene rs2976392 polymorphism was associated with an increased risk of NAFLD in north and south group population,additive model could reduce risk of NAFLD in south group population(OR=0.864(0.796,0.938)P<0.05).Conclusion:(1)Except additive model,allele model,dominant gene model,recessive model and codominant gene model all showed that the PNPLA3 gene rs2976392 polymorphism was associated with an increased risk of NAFLD in east-asain population.In additive model rs738409 polymorphism may reduce risk of NAFLD.(2)In additive model,rs738409 polymorphism could reduce risk of NAFLD in chinese group,adult group,south chinese group.
