Objective:To study the dynamic changes of brain functional states in insomnia patients and age-matched and sex-matched healthy controls by means of EEG microstates analysis,to determine the brain topological structure and temporal characteristics of sub-second brain activity,and to explore the relationship between various EEG microstates parameters and clinical scales and cognitive function.Research methods:Basic information collection,sleep quality scale and cognitive function assessment were carried out for the chronic insomnia group and the normal control group.After that,standard polysomnography monitoring was conducted overnight,and 2-5 minutes of sleep fragments of W,N1,N2,N3 and R stages were extracted for microstate analysis.Microstate time parameters(mean duration,Occurrence and Contribution)were statistically analyzed and correlated with cognitive function.Results:1.In general clinical data and scale evaluation,scores of PSQI,ISI,HAMA and HAMD-17 in insomnia group were higher than those in normal control group.2.The comparison of PSG sleep parameters between the insomnia group and the normal group showed that the total sleep time,sleep efficiency,sleep latency,wake time after sleep,the proportion of N2 stage increased,the proportion of N3 stage decreased in the insomnia group.3.There were no significant differences in overall memory quotient,long-term memory,short-term memory and instantaneous memory between the insomnia group and the normal group,but there were differences in individual memory.In long-term memory,the number sequence relationship was significantly different,and the insomnia group had lower scores from 1 to 100.In short-term memory,the picture recall ability of insomnia group was weak.In instantaneous memory,the insomnia group had fewer digits to recite backwards.In the insomnia group,attention was impaired and executive function was not abnormal.4.In comparison of eeg microstates,it was found that the dominance of microstate C in N3 and R phases was weakened,and its duration,occurrence frequency and coverage rate were all low.5.There was no correlation between general clinical features and microstate parameters in insomnia group.The proportion of N2 was negatively correlated with the duration of N3 microstate C,and the duration of N3 microstate C was positively correlated with reverse digit span test.Conclusion:The sleep structure of chronic insomnia patients is disordered,and the microstates parameters of waking state are not significantly different from those of normal people.The dominance of microstate C in N3 and R stages of chronic insomnia was weakened,and the cognitive function was correlated with the duration of N3 microstate C.
