Objective: To establish a rat model of allergic rhinitis(AR)due to lung and spleen Qi deficiency syndrome(LSQDS),and to intervene with Qichai decoction,to observe the expression levels of Th17/Treg cell-related factors Foxp3 m RNA,RORγtm RNA and serum IL-17 in rats.The effect of Qichai decoction on the balance of Th17/Treg cells in allergic rats with lung and spleen Qi deficiency syndromeMethods: 1.Select 24 SD rats,half male and half male,and randomly divide them into blank control group,lung-spleen-spleen-deficiency AR group,and AR group;lung-spleen-spleen-deficiency AR group rats were given smoky+ rhubarb decoction by gavage + OVA sensitization For modeling,the rats in AR group were sensitized by OVA.,and the blank control group was given the same dose of normal saline.The general condition of the rats was observed,and then the LSQDS rat model was evaluated according to the objective indicators of body weight,and weight-bearing swimming time;the pathological changes of nasal mucosa were observed,and the serum Ig E level and behavior were detected by enzyme-linked immunosorbent assay(ELISA)behavioral score,were used to evaluate the AR rat model of LSQDS.2.The LSQDS AR rat model was replicated,and the traditional Chinese medicine concentrate of Qichai decoction(13.75g/kg.d)was intragastrically administered for 4 weeks to observe the general behavior,behavioral scores and pathological changes of the nasal mucosa of the rats,and detected by ELISA.Serum IL-17 level,detected by real-time quantitative PCR in nasal mucosa of ratsResults: 1.The model is established successfully.The behavioral scores of AR group and AR group with LSQDS were both greater than 5 points;under light microscope,the structure of nasal mucosa epithelial cells of rats was severely damaged,the mucosal layer was thickened,glands increased,and a large number of inflammatory cells were infiltrated;AR group with LSQDS The nasal mucosa injury was more serious in AR group.Compared with the blank control group,serum Ig E levels in AR group and AR group with LSQDS were significantly higher(P<0.05);the AR group with LSQDS was more obvious.In addition,the AR group with LSQDS showed symptoms of lung and spleen qi deficiency such as shortness of breath,slow activity,shortness of breath,dry and dull hair,mental fatigue,decreased appetite,huddled together,and loose stools.At the same time,body weight and weight-bearing swimming time were significantly decreased.Compared with the blank control group and AR group,the difference was statistically significant(P<0.05),there was no significant difference in the above symptoms and indicators between the blank control group and AR group.2.Compared with the model control group,the level of serum IL-17 and the expression of RORγt m RNA in the nasal mucosa of the traditional Chinese medicine Qichai decoction group were decreased,and the difference was statistically significant(P<0.05);the expression of Foxp3 m RNA in the nasal mucosa was significantly increased,and the difference was Statistical significance(P<0.05).Conclusion: 1.Smoking + rhubarb decoction gavage + controlled diet +ovalbumin sensitization method can establish the AR rat model of lung and spleen Qi deficiency syndrome.2.Chinese medicine Qichai decoction can reduce the level of serum IL-17,the expression of RORγtm RNA in the nasal mucosa,and increase the expression of Foxp3 m RNA in the nasal mucosa after intervention in AR rats with LSQDS;The infiltration of inflammatory cells is reduced,and its mechanism may be related to down-regulating the expression of RORγt,up-regulating the expression of Foxp3,promoting the generation of Treg cells,inhibiting the differentiation of Th17 cells and the secretion of IL-17,thereby correcting the imbalance of Th17/Treg cells.
