Study On Protective Effect And Mechanism Of Mongolian Medicine Qing Gan Jiu Wei Powder On Hepatic Fibrosis Rats

Objective: 1.To study the protective effect of QGJWS on rats with hepatic fibrosis by comparing liver pathological changes and serum biochemical indexes before and after treatment,and to detect ERK pathway-related proteins by molecular biology techniques.2.To find the target of Mongolian medicine QGJWS regulating ERK signaling pathway in the treatment of liver fibrosis,so as to provide experimental basis for the treatment of liver fibrosis with this drug.Methods: In this study,48 SD rats were randomly assigned to model group,normal group,positive drug group(silybin)and Mongolian medicine QGJWS group(high,medium and low).After modeling,corresponding drugs were administered intragastric therapy respectively.After the experiment,all rats were sacrificed and blood and liver tissue samples were obtained to compare serum liver biochemical indexes of rats in each group.HE staining and Tianwolf scarlet staining were used to compare liver pathological changes of rats in each group after treatment.The expression of COLIm RNA,EGFR m RNA and COLI,α-SMA,EGFR and ERK related proteins in liver tissues of rats were detected by immunohistochemistry,real-time PCR and Western blot.To evaluate and analyze the therapeutic effect of Qinggan Jiuwei Powder on hepatic fibrosis in rats and its effect on ERK pathway-related proteins.Results: The general mental state of QGJWS rats was improved,liver damage was relieved,but the levels of serum transaminase and total cholesterol were significantly reduced.The experimental results showed that compared with the normal group,the expression of COLIm RNA,EGFR m RNA and COLI,α-SMA,EGFR and ERK related proteins in the model group were significantly increased.Compared with model group,the expression levels of COLIm RNA,EGFR m RNA and COLI,α-SMA,EGFR and ERK related proteins in Mongolian medicine groups were significantly decreased.Conclusion:(1)QGJWS can significantly improve the overall state of rats,reduce the liver biochemical indexes of rats with liver fibrosis,and significantly reverse the pathological manifestations of liver tissue,so it can be inferred that qinggan Jiuwei Powder has a therapeutic effect on liver fibrosis.(2)QGJWS could effectively control the expression levels of COLIm RNA,EGFR m RNA and COLI,α-SMA,EGFR and ERK-related proteins in rat liver fibrosis tissues,suggesting that the formula might regulate downstream Ras/RAF/MEK/ERK pathway through EGFR,and then reverse ECM deposition.In order to achieve the effect of treating liver fibrosis.
