Objective To study the effect of apolipoprotein A2(APOA2)gene expression on the replication and expression of hepatitis B virus(HBV).The correlation between APOA2 and the progression of CHB was analyzed.Methods Overexpression plasmid and si RNA were transfected into HepG2.2.15 cells respectively.Culture for 72 hours.RT-q PCR,Western bolt and ELISA were used to detect APOA2,HBV-DNA,HBs Ag,HBe Ag and HBc Ag,which inside and out inside the cell.Clinical specimens were collected according to the inclusion criteria.Normal control group(group N),chronic HBV carrying group(CHB1),HBe Ag positive CHB group(CHB2)and hepatitis B cirrhosis decompensated group(CHB3),each 50 cases were taken as the research object.In addition,patients with HBV-DNA content greater than 10~7IU/ml for the first time were taken as the research object.The patients were divided into groups according to the decrease of HBV-DNA content before treatment and 6 months after treatment.Group 1 was HBV-DNA≥10~5IU/ml,group 2 was HBV-DNA≥10~2IU/ml,and group 3 was lower than the detection line.The contents of apoa2 and HBV-DNA in fasting venous blood of the above patients were detected by immunoprojection turbidimetry and RT-QPCR.The test results of each group were compared by t-test,and the correlation between apoa2 content and CHB disease progression was analyzed.Results APOA2 m RNA and protein levels in HepG2.2.15 cells were significantly lower than those in HepG2 cells(P<0.05);When APOA2 gene was overexpressed in HepG2.2.15 cells,the contents of HBV-DNA,HBs Ag,HBe Ag and HBc Ag in and out of HepG cells decreased significantly(P<0.05);The above indexes were significantly increased when apoa2 gene was silenced(P<0.05);Apoa2 decreased gradually among N group,CHB1 group,chb2 group and chb3 group,and there was significant difference between the two groups(P<0.05);On the contrary,the content of apoa2 showed an increasing trend among group 1,group 2 and group 3,and there was significant difference between groups(P<0.05).Conclusion APOA2 can inhibit HBV-DNA replication and expression.It plays a role in inhibiting the progression of CHB caused by HBV.
