Objectives:Hyperhomocysteinemia(HHcy)is an easily neglected disease,which can be hidden in the body for many years without signs or symptoms.We want to explore the causes of HHcy,the correlation of HHcy with micronutrients,lifestyle,dietary habits and HHcy related genes,preventing the occurrence of HHcy.Methods:800 subjects(M/F,403/397)by plasma Hcy levels were divided into normal Hcy and HHcy group,using the questionnaire method to compare the basic characteristics of participants,and randomly selected 90 participants completed our own detailed questionnaires about lifestyle and eating habits,and randomly selected203 cases of patients using Real-Time PCR fluorescent probe method to detect folic acid related gene polymorphism,including MTHFR C677 T,MTHFR A1298 C and MTRR A66 G three sites.Micronutrients,including blood vitamin D(25-OH-D3),zinc,iron,calcium,magnesium,lead,copper,alkaline phosphatase,folic acid and vitamin A and vitamin B12,were detected by mass spectrometry.Blood biochemistry includes blood Hcy and blood uric acid.The statistical analysis methods were chi-square test,analysis of variance,logistic regression,etc.The trial was registered with trial number CHICTR1900025136.Results:1.42.66% of HHcy patients were aged between 10 and 18 years.2.Compared with normal group,HHcy group of patients with highezinc(zinc),vitamin A,Uric acid(Uric)level(P(27)0.05),and lower plasma total 25-hydroxy-vitamin D3(25-OH-D3),vitamin B12(B12),folic acid(Fol)level(P(27)0.05).3.Regular drinking of a small amount of coffee,tea and soda as well as the habit of daily supplement of complex B group are the protective factors for HHcy,while the pressure of entering school,staying up late,depression,and low intake of vegetables and fruits are the risk factors for HHcy.4.HHcy detection rate was 17.88%(143/800),only MTHFR 677 TTmutation affect blood levels of Hcy(P(27)0.05),while the MTHFR1298 CC and MTRR 66 GG had no significant effects on the blood Hcy concentration(P(29)0.05).5.Multivariate logistic regression showed that plasma zinc level increased(Exp(B)= 1.042;95% CI,1.000-1.086),elevated plasma uric acid(Exp(B)= 1.014;95% CI,1.008-1.019),lower vitamin B12 level(Exp(B)=0.993;95%CI,0.989-0.997),and low folate level(Exp(B)= 0.772;95%CI,0.657-0.907),and MTHFR C677 T polymorphism 677 TT genotype(Exp(B)=2.923;95%CI,1.399-6.107)was positively correlated with serum Hcy level.Conclusion: Pay more attention to students who were facing great pressure,because they are very likely to be potential HHcy patientswith high levels of uric acid and zinc and anxiety.For the population whose genotype test result was MTHFR 677 TT,attention should be paid to the deficiency of related nutrients,timely improvement of lifestyle,and use of nutritional factors to correct the influence of genes on the disease. |
