| Tinea pedis is a fungal skin disease with the highest incidence in China.Among the pathogenic fungi causing tinea pedis,Trichophyton rubrum is the main pathogenic fungus,accounting for about 80%in clinical tests.Therefore,in this study,Trichophyton rubrum was taken as the model,and the efficacy of traditional Chinese medicine against Trichophyton rubrum was investigated according to the standards of Clinical and Laboratory Standards Institute(CLSI)with the minimum inhibitory concentration MIC as the evaluation index.Screening of the external use herbs have antifungal effect,found Charred Radix Sanguisorbae for Trichophyton rubrum has good bacteriostasis(MIC=64 mu g/mL),and no reports of Charred Radix Sanguisorbae inhibition of Trichophyton rubrum,so this study selectively Charred Radix Sanguisorbae as the main research object,explore Charred Radix Sanguisorbae in the main effective component and function mechanism in the treatment of tinea.Radix Sanguisorbae is the dried root or rhizome of Sanguisorbal(Rosaceae).The Charred Radix Sanguisorbae is the processed medicinal material of Radix Sanguisorbae.The changes of chemical constituents of Radix Sanguisorbae after processing into Charred Radix Sanguisorbae are relatively complex.Although there are systematic studies on the constituents of Radix Sanguisorbae in domestic and foreign literatures at present,systematic analysis of chemical constituents of Charred Radix Sanguisorbae is lacking.At present,the pharmacodynamic studies on the preparation of Charred Radix Sanguisorbae are mainly focused on hemostasis,and there are few studies on bacteriostasis.Studies have found that the bacteriostatic effect of Charred Radix Sanguisorbae is better than Radix Sanguisorbae.To study the composition difference of Radix Sanguisorbae before and after processing and the material basis of its antibacterial action,HPLC-Q/TOF-MS/MS method was used to analyze and study the medicinal materials.It was found that the content of tannins in Charred Radix Sanguisorbae was significantly increased after processing,especially the content of ellagic acid was significantly increased,which may be the material basis of Charred Radix Sanguisorbae better inhibitory effect on Trichophyton rubrum.To study the bacteriostasis mechanism of Charred Radix Sanguisorbae,it was found that the extracts of Charred Radix Sanguisorbae could inhibit the normal growth of biofilm,cell membrane and cell wall of Trichophyton rubrum.The cytotoxicity was decreased after processing.It also has good inhibitory effect on other common fungi of skin diseases.It is a good drug for the treatment of skin diseases caused by skin fungi.Taking gallic acid and ellagic acid as the index components,it was found that the optimum condition for frying Charred Radix Sanguisorbae was processed at 290℃ for 6 min.In the end,the extract of Charred Radix Sanguisorbae was prepared into Charred Radix Sanguisorbae gel,and its formulation was optimized and its quality was evaluated.This study started from the screening of medicinal materials,with Trichophyton rubrum as the model,and found a new medicinal material for the treatment of tinea pedis.The material basis and mechanism of the inhibition effect of Charred Radix Sanguisorbae on Trichophyton rubrum were studied.Finally,the gel was developed to enlarge the application range of Charred Radix Sanguisorbae. |
