The dried stem of Schisandra chinensis which belongs to Magnoliaceae can be called Zanthoxylum bungeanum.It is mainly used as cooking seasoning,and as clinical drug,the research of large-scale production is relatively less.It was found that ethanol extract of Schisandra chinensis stem could effectively prevent liver injury in mice.Because it contains lignan,a chemical component similar to fruit.Lignan is low molecular secondary metabolites which is composed of Biphenyl cyclooctadiene.It is the main active substances of Schisandra chinensis fruit.Lignan has many biological activities.In recent years,with the increasing development and utilization of Schisandra chinensis stem,large-scale artificial planting has been carried out in Northeast China.Therefore,the development and utilization of Schisandra chinensis stem,whether it can be used as a substitute for its fruit,and even characteristic drugs are important.In this experiment,Schisandra chinensis was selected as the research object,The method of detecting five representative lignans in Schisandra chinensis by HPLC was explored,such as schizandrin A,schizandrin B,schizandrin C,schizandrol A,schizandrol B.The linear ranges of schizandrin A,schizandrin C,schizandrol A,and schizandrin B,schizandrol B were 0.02-0.2mg/mL and 0.05-0.4mg/mL,respectively.The recoveries were 91.19%,88.59%,90.58%,92.75%and 92.92%,respectively;The RSD of precision test and reproducibility test were less than 3%,and the RSD of stability test was less than 2%.The results showed that the method is rapid,sensitive,specific and accurate.The contents of five components were used as evaluation indexes.The effects of solid-liquid ratio,extraction times,extraction time and ethanol concentration on the extraction rate of total lignans were investigated by single factor experiment.The extraction process of total lignans was optimized by by box-Behnken.The optimum conditions were as follows:solid-liquid ratio1:10,extraction 3times,extraction 5h and 95%ethanol concentration.The verifying test results showed that the extraction amounts of schizandrol B,schisandrin B and total lignans were 15.2987mg/g,10.8967mg/g and 31.7395mg/g respectively.Purify with AB-8macroporous resin.The purification process was as follows:sample concentration was 1.5mg/mL,adsorption rate was 2BV/h,sample volume was 6BV,95%ethanol was used for elution,elution rate was 2BV/h,eluent dosage was 8BV.The average purity of total lignans was 59.7%.Finally,the scavenging rates of total lignans on hydroxyl radical and superoxide anion radical were studied.The results showed that the total lignans had scavenging ability to hydroxyl radical and superoxide anion radical,and the scavenging ability increased with the increasing of total lignans concentration,At the concentration of 0.2mg/mL,the antioxidant capacity of Schisandra chinensis lignan solid complex was 1/4~1/5 of VC,indicated that it had certain antioxidant capacity.In this study,the quantitative analytical methods of total lignans of Schisandra chinensis was established.The extraction process and purification method of total lignans were determined.The composition of lignans of Schisandra chinensis stem was verified.The waste parts of traditional Chinese medicine can be reused.This study provides some reference value for the large-scale production and functional research of Schisandra chinensis.Finally,the total antioxidant activity and superoxide anion scavenging free radical test showed that the total lignans solid of Schisandra chinensis had certain antioxidant effect.
