| Objective:To investigate the effect of Coix Lacrymajobi Seed Oil(CLSO)on the atopic dermatitis(AD)model in mice induced by 2,4-dinitrochlorobenzene(DNCB);and study the regulation mechanisms of CLSO on the expression of cornified envelope(CE)components,such as the filaggrin(FLG),involucrin(IVL),loricrin(LOR),and the upstream targets—aryl hydrocarbon receptor(AHR)and Ovo-like 1(OVOL1).Method:Divided BALB/c mice stochastically into four groups:Control(the blank control group),Model(the model group),CLSO(the CLSO treatment group)and Solvent(the CLSO solvent treatment group).The changes of ear thickness,scratching numbers and scoring atopic dermatitis index(SCORAD)of each group were recorded.After the intervention,fluorescent dye penetrat ion test,H&E staining and toluidine blue staining were performed,ear thickness was measured.Real-time PCR and Western blot were used to detect the expression of CE related molecules(including filaggrin(FLG),loricrin(LOR),involucrin(IVL)),as well as the upstream regulatory target of aryl hydrocarbon receptor(AHR)and the susceptibility gene Ovo-like 1(OVOL1).Result:After intervention,thickness of ear,numbers of scratching,SCORAD and epidermal thickness in the CLSO treatment group were significantly reduced,which entered the scope of mild dermatitis.H&E staining and toluidine blue staining also confirmed the therapeutic effect of CLSO.After treatment with CLSO,the expressions of mRNA and protein of FLG,IVL and LOR in the skin of AD mice were significantly up-regulated.In addition,the mRNA and protein expressions of AHR and OVOL1 in the skin of AD mice treated with CLSO were significantly upregulated.ConclusionCLSO can significantly improve the inflammation of AD mice and repair the skin mechanical barrier function.Its mechanism may be to activate AHR,up regulate OVOL1,and then promote the expression of FLG,LOR,IVL.so as to repair the damaged skin barrier. |
