About 15%of couples have problems with infertility in the world,and half of these cases are due to male causes.However,oligoasthenospermia which is caused by reactive oxygen species(ROS),and eventually leads to sperm dysfunction,is an important cause of male infertility,accounting for 25%-40%of male infertility patients.The prevailing treatment for oligoasthenospermia is antioxidant supplementation.However,it is induced by a variety of factors,not a relatively independent disease,and the pathogenesis of the disease has not yet been fully elucidated.Therefore,there are some limitations in the treatment of western medicine and traditional Chinese medicine(TCM),mainly reflected in the treatment of clinical curative effect is not ideal,didn’t significantly change the status quo of male infertility.In recent years,assisted reproductive technology(ART),as a new treatment method,has improved the pregnancy rate of oligoasthenospermia patients,but the quality of sperm has not been fundamentally improved.The search for more effective antioxidants is an important way to solve the fundamental problem of improving sperm quality.The polysaccharides,proteins,astaxanthin,β-carotene,phenolic compounds and other active substances in algal extracts have been proved to have antioxidant activity,and have the advantages of high efficiency,non-toxicity,high quality and low price.Research on the potential effects of algae antioxidant compounds in the treatment of male infertility will not only help to clarify the mechanism of improving sperm activity,but also have a broad application prospect.In this paper,we selected algal powder,polysaccharide and pigment extracts from common algae with potential antioxidant activity.There are 10 different kinds of algae extracts,including the powder of A.platensis,10%extracts of A.platensis,30%extracts of A.platensis,10%polysaccharide of Saccharina japonica,30%polysaccharide of Saccharina japonica A,30%polysaccharide of Saccharina japonica B,5%astaxanthin,5%fucoxanthin and the powder of Chlorella pyrenoidosa.The scavenging ability of hydroxyl radical,superoxide anion,DPPH radical and ABTS radical was measured and compared.Combining with the physical and chemical properties of 10 kinds of algal extracts,the mechanism of their antioxidant activity in vitro was analyzed.At the same time,the therapeutic effect of TCM combined with algal extract on asthenospermia model rats was studied through animal experiments,in order to explore the mechanism of algae antioxidant substances to improve sperm activity.The main results and conclusions are as follows:1.The results of antioxidant activity in vitro showed that the scavenging ability of A.platensis powder is the strongest for hydroxyl radicals,and the IC50 is 0.01±0.01 mg/mL;for superoxide anions,5%fucoxanthin have the strongest scavenging ability,with IC50 value of 35.03±2.89 mg/mL;for DPPH free radical,5%astaxanthin have the strongest scavenging activity with IC50 value of 4.55±0.14 mg/mL;for ABTS radical,30%polysaccharide of Saccharina japonica B have the strongest scavenging activity with the IC50 value of 4.67 ±0.13 mg/mL.2.Combining with the physicochemical properties of 10 kinds of algae extracts,the antioxidant activity of the extracts was analyzed.The results showed a significant negative correlation between OH·clearance and sugar content(Pearson,P<0.05).There was a significant positive correlation between ABTS+·clearance and sugar content(Pearson,P<0.05).This indicates that the presence of sugar components plays a certain role in their antioxidant properties.3.The model of asthenospermia was created by different doses of ORN and the animal experiment of algae combined with traditional Chinese medicine was carried out.The results showed that,600 mg/kg dose of ORN is more appropriate tomake asthenospermia modle male SD rats.In all the treatment groups,the therapeutic effect of Chinese medicine combined with 30%polysaccharide of Saccharina japonica B on asthenospermia model rats is more significant.4.Low TCM and high polysaccharide of Saccharina japonica group had significant therapeutic effect on asthenospermia model rats,compared with the sperm concentration(22.7±4.32×106/mL)、sperm motility(8.75 ± 9.01%)、MDA level(18.10±6.08 nmol/mg prot)and SOD activity(126.42±5.67 U/mg prot)in model group,sperm concentration(43.8 × 8.4 × 106/mL)and sperm motility(58.08±11.88%)were significantly increased by 93.0%and 102.0%respectively,the MDA level(7.49±3.59 nmol/mg prot)of epididymal tissue was significantly decreased by 586%,SOD activity(136.32±14.80 U/mg prot)was increased.5.The combination of algae extracts antioxidant activity and the therapeutic effect of TCM in asthenospermia,has potential therapeutic effects in 600 mg/kg ORN-induced asthenospermia modle rats.It has a protective effect on sperm,improved the model of rat sperm concentration and sperm motility,and improved the level of the epididymis tissue antioxidant,decreased the OSs injury of epididymis tissue.
