| Colonic disease is one of the most common human diseases.Colitis and colorectal cancer bring a lot of burden to human life.Meanwhile,there is still a lot of challenges for the research of colonic diseases.The causes of colitis and colon cancer remain elusive,but the search for them never stops.Current studies have proved that hydrogen sulfide,the third largest gas signaling molecule,plays an essential physiological role in the intestinal tract.Hydrogen sulfide is present abundantly in the gut and provides the energy needed for intestinal homeostasis.In addition,previous studies have shown that the expression of hydrogen sulfide synthase is increased in colorectal tumor tissues,and hydrogen sulfide synthase CBS has been reported to promote the occurrence and development of colorectal cancer.CTH,as a hydrogen sulfide synthase,also plays a crucial role in colorectal cancer,but the specific mechanism is still not clear.Therefore,in this study,we first analyzed the expression level of CTH protein in human colorectal cancer samples.It was clear that the expression level of CTH was significantly up-regulated in human colorectal cancer compared with paracancerous tissues.At the same time,we proved that down-regulation of CTH expression inhibits the cell growth,colony formation,migration and invasion ability of colorectal cancer cells.Later,we demonstrated that CTH down-regulation inhibits cell proliferation through cycle arrest,but the modes of cell cycle arrest were different in different cell lines.Finally,we verified the inhibitory effect of CTH knockout on colorectal cancer in vivo through AOM-DSS induced colitis-associated mouse CRC model.In conclusion,our study showed that CTH is highly expressed in colorectal cancer and plays a promoting role in the initiation and development of colorectal cancer. |
