Keloid is a skin fibroproliferative disease currently has no uniformly satisfactory treatment.Clinically,keloid rarely regresses over time,often grows beyond the borders of the original wounds and continuously invades the surrounding normal tissues.All of these reflect the trait of tumor tendency of keloids.The bulky keloid tissues not only cause unsightly appearance and local irritating symptoms but also exert huge psychological pressure on patients.Keloid is comprised of actively proliferating and collagen overproducing fibroblasts,but the mechanism of uncontrol proliferation and invasive growth of keloid is still unclear,which is the bottleneck of targeted clinical treatment.La related protein 6(LARP6)is an RNA binding protein,which can regulate collagen synthesis in fibroblasts and tumor cell proliferation and invasion.To explore LARP6’s likely functions in keloid pathogenesis,we performed immunohistochemistry staining on human keloid tissues and discovered markedly upregulation of LARP6 expression in lesion fibroblasts compared to that of normal skin and hypertrophic scar tissues.In addition,the keloid tissue derived fibroblasts(KF)showed constitutive upregulation of LARP6 expression,as well as significantly upregulated mRNA and protein expressions of type Ⅰ collagen,enhanced cell proliferation and invasive behavior in cell culture system.Intriguingly,LARP6 known by a targeting siRNA significantly inhibited type Ⅰ collagen expression,proliferation and invasion capability of KF relative to the that of normal skin derived fibroblasts or KF that were transfected with a scrambled control siRNA.In conclusion,the abnormally upregulated expression of LARP6 in fibroblasts may play an important role in the growth and invasive behavior of keloid lesions.
