Supercritical Fluid Chromatography(SFC)is an environmentally friendly chromatographic technology.Supercritical fluids have the advantages of the inherent gas-like low viscosity and high diffusivity and liquid-like similar density,so SFC exhibits some interesting features,such as high separation efficiency,short analysis time,less organic solvents consumption and environmental friendliness.SFC has a good application prospect in the fields of chromatographic separation and analysis.Tandem Mass spectrometry has been widely applied to the fields of food safety,pharmacokinetics analysis,medicines and environmental monitoring owing to the properties of high sensitivity,high selectivity and rapid analysis.In this work,we have successfully developed two methods,SFC-MS/MS and SFC-LC-FLD,which have been applied to the analysis of triterpenic acids in Osmanthus fragrans fruits and on-line pretreatment of benzo(a)pyrene(Ba P)in the edible vegetable oil,respectively.The main contents of this work include three parts.In the first part,a simple and sensitive UHPLC-MS/MS method was developed and applied to detect ten triterpenic acids in Osmanthus fragrans fruits and we compared the content difference which originate from four provinces.The results indicated that oleanolic and ursolic acid predominate in all samples,which is consistent with the literature data.Among the minor components,arjunolic acid was detected in samples from Wuhan and Jinhua while in the two samples from Changsha and one from Jinhua,corosolic acid was absent.These results demonstrated that origin areas of traditional Chinese medicines should be considered when medicines are used for treatment.In the second part,we developed a SFC-MS/MS method for the analysis of triterpenic acids in Osmanthus fragrans fruits.Compared with the UHPLC-MS/MS method described in the first part,this method has the advantages of 5-10 fold higher sensitivity and shorter analysis time.What’s more,the quantitative results between these two methods were consistent,which proved two accurate methods and the SFC-MS/MS was superior.In the third part,we developed a SFC-LC-FLD method for on-line pretreatment and separation of benzo(a)pyrene in the edible vegetable oil.The method was simple without any fussy preparation procedure and has properties of good repeatability and clean-up effects.Twenty-three samples of different kinds of vegetable oil were analyzed and all of them were not above the limitation of national standard.Our work indicates the SFC-LC-FLD method are a promising pretreatment technique in other lipid-soluable samples.
