In this study,nineteen compounds from ethyl acetate fractions of Cercis chinensis Bunge were isolated by methanol immersion extraction,combined with silica gel,Sephadex LH-20,macroreticular resin column chromatography and recrystallization.The chemical structures of isolated compounds are elucidated by NMR.The isolated compounds were identified as vanillic acid(1),β-sitosterol(2),2-methoxyhydroquinone(3),isoferulic acid(4),p-hydroxybenzoic acid(5),(1,5-di-p-hydroxyphenyl)-1,4-pentadien-3-one(6),kaempferol(7),luteolin(8),quercetin(9),apigenin(10),gallic acid(11),protocatechuic acid(12),kaempferol-3-O-(α-L-rhamnopyranoside)(13),isorhamnetin-3-O-(β-D-glucoside)(14),quercetin-3-O-(α-L-rhamnopyranoside)(15),myricetin-3-O-(α-L-rhamnopyranoside)(16),kaempferol(17),1,3-dihydroxyindole(18)and 7-hydroxycoumarin(19)on the basis of NMR spectroscopic analysis.Among them,compounds5、13、18、19were isolated from the plant for the first time.Evaluated of the inhibitory effect of isolated compounds on HIF-1α activation in Hep3B cells by Luciferase reporter assay,and cell viability was measured by MTT assay.As a result,compound 4 and 18 exhibited significant HIF-1α inhibitory activities.Compound 2 had strong cytotoxicity to HIF-la from MTT test results.
