Objective:Using protein microarray technology to screen serum markers of patients with psoriasis and explore the significance of their expression,so as to provide new methods and ideas for clarifying the pathogenesis of psoriasis and developing clinical targeted drugs.Methods:1.Collect the serum of patients who were treated in the Affiliated Hospital of yanbian University from June 2018 to December 2019(30 patients with psoriasis and 20 patients from the health examination department).According to the clinical manifestations,the skin lesion and severity index(PASI)of psoriasis was scored,and the emotional state and mental health level of patients with psoriasis were assessed by the mental stress questionnaire(current state and normal state).2.Using Protein microarray technology to detect the expression level of serum protein to be tested in psoriasis patients and normal control group,analyze the data and screen out the differentially expressed proteins.3.Analyzing the correlation between the expression level of differential proteins and the psoriasis patients’ PASI score and mental stress questionnaire score.4.Evaluation of the diagnostic value of differential proteins and their combinations.Results:1.The results of Protein microarray technology showed that among the detected proteins,the expression levels of GDF-15,DPP4 and ANGPTL3 in the serum of psoriasis patients group compared with the normal control group increased,the difference was statistically significant(P<0.05).2.The expression level of DPP4 was negatively correlated with PASI score(r=-0.529,P<0.05).The expression level of GDF-15 was positively correlated with the score of the mental stress questionnaire in the usual state(r=0.661,P<0.001);the expression level of ANGPTL3 was positively correlated with the score of the mental stress questionnaire in the current state(rs=0.595,P<0.05).3.The area under ROC curve of GDF-15,DPP4 and ANGPTL3 is 0.730,0.665 and 0.712,respectively,which have diagnostic value for psoriasis.The area under the ROC curve of GDF-15 combined with DPP4 is 0.803,which has higher diagnostic value.Conclusion:1.GDF-15,DPP4 and ANGPTL3 are related to the occurrence and development of psoriasis,and the serum expression level of DPP4 has a negatively correlated with the severity of psoriasis,which can be used as an indicator to judge the severity of the disease.2.GDF-15,DPP4 and ANGPTL3 have certain diagnostic value for psoriasis,and GDF-15 combined with DPP4 has higher diagnostic value for psoriasis. |
