Astringent medicine is a kind of important medicine widely used in the clinical treatment of body fluid dissipation and slippage and other diseases.As the representative medicine of astriction,Schisandrae Chinensis Fructus is sour,sweet and warm in nature,belongs to the lung meridians,heart meridians and kidney meridians.It has the functions inducing astringency,replenishing and promoting production of body fluid and tonifying the kidney to relieve mental strain.It is often used in chronic cough and asthma,enuresis and frequent micturition,sponteneous sweating,night sweating and other body fluid dissipation and slippage disease.However,modern pharmacological studies show that the activities of Schisandrae Chinensis Fructus and other astringent and astringent drugs are mostly concentrated in anti-cancer,lipidlowering,liver-protecting and other aspects,which are significantly different from the traditional treatment.The main reason for this difference lies in the lack of scientific characterization,accurate understanding and appropriate research models of traditional chinese medicine theories such as astriction and body fluid dissipation and slippage.Based on the absorption characteristics of water and nutrients by lumen epithelial cells mediated by sodium potassium ATPase,this study found that the system has a high similarity with the theory of body fluid of traditional Chinese medicine.Therefore,the scientific hypothesis that NKA-mediated absorption system of lumen epithelial cells may be the key mechanism of astriction of traditional chinese medicine is proposed.To verify this hypothesis,a quantitative method of NKA based on ICP-MS was established.This method has the advantages of high sensitivity and stability compared with conventional colorimetry,and can avoid isotope contamination.Using this quantitative method,it was found that astringent and astringent drugs could specifically activate NKA activity in lung and intestinal epithelial cells,and promote cell absorption of water and glucose,but not astringent and astringent drugs.Schisandrae Chinensis Fructus as a representative drug,found that raw and vinegar Schisandrae Chinensis Fructus can significantly activate NKA activity of intestinal epithelial cells,and protect intestinal epithelial cells NKA function is suppressed,but there is no significant difference between raw and vinegar Schisandrae Chinensis Fructus.Further studies showed that Schisandrae Chinensis Fructus lignans had NKA protective effect on lung and intestinal epithelial cells,but there was no significant difference in NKA activity between raw Schisandrae Chinensis Fructus and vinegar Schisandrae Chinensis Fructus because of the low total content of lignans in raw Schisandrae Chinensis Fructus and vinegar Schisandrae Chinensis Fructus.Mitochondria are the main organelles of NKA.Based on the mitochondrial damage model induced by rotenone,we found that Schisandrae Chinensis Fructus lignans could significantly inhibit the decrease of MMP and the increase of ROS in mitochondrial dysfunction,and some Schisandrae Chinensis Fructus lignans could inhibit the formation of ROS in normal cells.Therefore,protecting the function of mitochondria and providing enough energy for NKA in epithelial cell membrane may be the common and important biological mechanism of Schisandrae Chinensis Fructus as a representative of astringent traditional chinese medicine.This study reveals the new mechanism of action of Schisandrae Chinensis Fructus and other astringent drugs,and provides a new theoretical basis for the rational use of these drugs in clinic.
