| Objective:To investigate the expression and function of Rab23 gene in papillary thyroid carcinoma.Methods:(1)The expression of Rab23 and miR-221、222 gene was examined in papillary thyroid carcinoma tissues and adjacent tissues of 38 patients by real time fluorescence quantitative PCR.(2)The expression of Rab23 and miR-221、222 gene was detected in papillary thyroid carcinoma cell lines(TPC-1,K1,BCPAP)and normal thyroid cell line(Nthyl)by qRT-PCR.(3)K1 cell was stably infected by Rab23-RNAi lentiviral vector in order to knock down the expression of Rab23 in K1 cell.(4)The effects of Rab23 knock-down on the abilities of proliferation and invasion of K1 cell were explored by MTT test,clone formation experiment,wound healing assay and Transwell experiment.Results:(1)The expression level of Rab23 gene in cancer tissues of papilary thyroid carcinoma was significantly lower than that in corresponding adjacent tissues(P<0.05).The expression of miR-221、222 in cancer tissues of papilary thyroid carcinoma was significantly higher than that in corresponding adjacent tissues(P<0.05).The correlation between them was not significant.(2)The expression of Rab23 in papillary thyroid carcinoma cell lines(TPC-1,K1 and BCPAP)was significantly lower than the expression in normal thyroid cells(Nthyl)(P<0.05).The expression of miR-221、222 in papillary thyroid carcinoma cell lines was significantly higher than the expression in normal thyroid cells.(3)K1 cell was successfully infected by Rab23-RNAi lentiviral vector and the expression of Rab23 in such cell was stably knocked down.(4)MTT test showed that Rab23 knock-down could effectively reduce the abilities of proliferation;clone formation experiment showed that Rab23 knock-down could suppress the clone formation of K1 cell;wound healing assay and Transwell experiment showed that Rab23 knock-down could effectively reduce the abilities of invasion and metastasis of K1 cell.In a word,indicateConclusion:The expression of Rab23 in papillary thyroid carcinoma was down-regulated,while the expression of miR-221、222 was up-regulated.The correlation between them was not significant.Preliminary results showed that these three genes were related to papillary thyroid carcinoma.In addition,knockdown of Rab23 can effectively inhibit the proliferation、invasion and metastasis of K1 cells.These results have preliminarily explored the function of Rab23 gene in papillary thyroid carcinoma,which is beneficial to further understanding the occurrence and development of papillary thyroid carcinoma. |
