Objectives To investigate the expression of Smad7 in oral squamous cell carcinoma(OSCC)and determine if Smad7 was associated with tumor cell proliferation and migration.Methods 1.Immunohistochemistry was used to detect Smad7 expression in oral squamous carcinoma tissues and corresponding normal tissue samples.2.Total protein was extracted from 4 OSCC oral squamous carcinoma tissues and corresponding normal tissue samples and the expression of Smad7 was detected by Western blot.3.CCK8 and Cell scratch assay were used to evaluate the effect of Smad7siRNA on proliferation and migration of OSCC cells.4.Western blotting and qRT-PCR were performed after Smad7siRNA interference in HN4 cells.Results 1.Nuclear and cytoplasmic expression of Smad7 were increased in primary OSCC tissue samples.2.Expression of Smad7 was associated with lymph node metastasis.3.Smad7 knockdown greatly suppressed cell proliferation but promoted tumor cell migration.4.Smad7 knockdown promoted N-cadherin and Vimentin expression and suppressed Ecadherin expression.Conclusions Smad7 expression in OSCC was associated with lymph nodal metastasis and Smad7 might play an essential role on the proliferation and migration ability of OSCC cells. |
