Objective:To explore the effect of Buyanghuanwu decoction(BYHWD)with different components and different doses of astragalus membranaceus on the neurogenesis and angiogenesis after focal cerebral ischemia in rats.Methods:Focal cerebral ischemia was induced by occlusion of the right middle cerebral artery for 90 min using the intraluminal filament model.BYHWD with different components(decoction、tonifying Qi、activating blood)and different doses of astragalus membranaceus(120 g、60 g、30 g and 15 g)and BrdU(50 mg/kg,ip)were treated on day 1 after ischemia once a day for 14 days.The modified neurological severity score(mNSS)and the corner test were used to evaluate sensorimotor function on 1,7,and 14 days after ischemia.BrdU/Nestin、BrdU/GFAP、BrdU/NeuN、BrdU/vWF positive cells were examined by double immunofluorescence,BDNF positive cells were examined by single immunofluorescence on 14 days after ischemia.Results:The effect of Buyanghuanwu decoction(BYHWD)with differentcomponents on the neurogenesis and angiogenesis after focal cerebral ischemia in rats:Compared with the model group,(1)BYHWD with different components(decoction and tonifying Qi)significantly ameliorated neurological dysfunction and reduced the number of right turn on 7 and 14 days after ischemia(P<0.05 or P<0.01).(2)the number of BrdU/Nestin positive cells in SVZ of decoction and tonifying Qi was significantly enhanced on 14 days after ischemia(P<0.05 or P<0.01).(3)BYHWD with different components(decoction)significantly enhanced the number of BrdU/GFAP positive cells(P<0.01),(4)the number of BrdU/NeuN positive cells in SVZ of BYHWD with different components(decoction)was significantly enhanced on 14 days after ischemia(P<0.01).(5)The number of BrdU/vWF positive cells in SVZ of BYHWD with different components(decoction)was significantly enhanced on 14 days after ischemia(P<0.01),(6)BYHWD with different components(decoction)enhanced the number of BDNF positive cells(P<0.01)on 14 days after ischemia.The effect of Buyanghuanwu decoction(BYHWD)with different doses of astragalus membranaceus on the neurogenesis and angiogenesis after focal cerebral ischemia in rats:Compared with the model group,(1)BYHWD with astragalus membranaceus(120 g and 60 g)significantly ameliorated neurological dysfunction and reduced the number of right turn on 7 and 14 days after ischemia(P<0.05 or P<0.01).(2)The number of BrdU/Nestin positive cells of the astragalus membranaceus(120 g and 60 g)in SVZ was significantly enhanced on 14 days after ischemia(P<0.01 or P<0.05).(3)The number of BrdU/GFAP positive cells of the astragalus membranaceus(120 g)in SVZ was significantly enhanced on 14 days after ischemia(P<0.01).(4)The number of BrdU/NeuN positive cells of the astragalus membranaceus(120 g)in SVZ was significantly enhanced on 14 days after ischemia(P<0.01).(5)The number of BrdU/vWF positive cells of the astragalus membranaceus(120 g)in SVZ was significantly enhanced on 14 days after ischemia(P<0.01).(6)The number of BDNF positive cells of the astragalus membranaceus(120 g and 60 g)in SVZ was significantly enhanced on 14 days after ischemia(P<0.01 or P<0.05).Conlusion:(1)The mechanism of BYHWD decoction promotes the function of the nerve probably associated with that it could promote neurogenesis and angiogenesis(2)The effect of BYHWD decoction with different components tells that the BYHWD of tonifying Qi promotes the neurogenesis and angiogenesis,the effect of BYHWD with different components(decoction)was the best one whether on neurogenesis or angiogenesis.(3)BYHWD with higher dose of astragalus membranaceus promotes the neurogenesis and angiogenesisimproves neurological function recovery following focal cerebral ischemia,the effect of 120 g astragalus membranaceus is better.
