Ginger lotus(Curcuma alismatifolia Gagnep.)is an ornamental flower with high market value,which is very popular in South China and Southeast Asia,and is the main flower of Buddhism.Meanwhile,its bulbs are rich in curcumin which has high medicinal value and is used as an effective Chinese medicine in the recent popular COVD-19 treatment for its anti-inflammatory,antipyretic and detoxifying effects.At present,most of the ginger rhododendrons in the market are propagated by bulbs,which is inefficient,easily infected with pests and diseases,and greatly consume human and land resources,causing great difficulties for their breeding.Reproduction efficiency can be greatly improved by tissue culture,and strong technical support can be provided for the molecular breeding.This experiment establishes an efficient ginger lotus fast propagation system and ginger lotus regeneration system to solve the problem of efficient multiplication of ginger lotus from two directions respectively,as well as to provide an important technical reserve for molecular breeding.The main findings were as follows.1.The best way to sterilize explant bulbs: pretreatment with carbendazim spraying material,obtaining bulb buds and soaking them in hand PPM 30 min,followed by rinsing in running water for 1 h.After removing the brown oxidized material on the surface,soaking in 75% alcohol for 30 s,rinsing in sterile water three times,soaking in ascorbic acid for 15 min after shaking,rinsing in sterile water three times,the highest sterility rate of explants could reach 66.8 %.2.The optimal medium for the proliferation of adventitious buds of sterile seedlings is:MS + 30 g/L sucrose + 2.4 g/L gel + 2 mg/L 6-BA + 0.1 mg/L NAA can obtain a proliferation coefficient of 8.2,and the clumps The growth state and growth potential of the buds are the best,and the proliferation coefficient will be expanded exponentially with continued subculture.3.Optimal medium for root induction of sterile seedlings: MS + 30 g/L sucrose + 2.4 g/L gel + 0.2 mg/L IBA,the best performance is achieved,the average number of proliferating roots reaches 15.4,and the average root number reaches 15.4.The length reaches 6.3cm.4.The best medium for callus induction of ginger and lotus: MS + 30 g/L sucrose + 2.4g/L gel + 1 mg/L 2,4-D,under this condition,the callus induction rate can reach 82.3%,and the callus activity is strong,the proliferation ability is strong,and the state is good.5.The callus induction experiment was carried out on the roots,stems,leaves and bulb buds of ginger lotus.The results showed that the roots and leaves could not induce callus.Bud-induced callus is the best.6.The best medium for inducing differentiation of ginger lotus: MS + 30 g/L sucrose +2.4 g/L gel + 2 mg/L KT + 0.2 mg/L NAA,and the differentiation efficiency is the highest under this condition.
