| The substitution of antibiotics with Chinese veterinary medicine to solve the problems of resistance and veterinary drug residues is a hot issue in the prevention and control of epidemics in breeding farms.Based on the patent obtained by the research group in the previous stage,this study investigates the chemical composition,quality control and quality standard of the compound of flavonoids and baicalin polysaccharides from Yantuo,with the aim of laying a foundation for the application and development of this medicine.In this experiment,15 batches of Chinese herbs Yantuo and baicalin were collected,and flavonoids from Yantuo and polysaccharides from baicalins were extracted by ethanol extraction and water extraction-precipitation respectively,and the yield was calculated;two extracts were mixed at a ratio of 1:1 to obtain the compound of flavonoids and baicalin polysaccharides;liquid chromatography-mass spectrometry was used to analyze and identify its chemical composition;a high-performance liquid chromatography method was established,and a fingerprint similarity evaluation system(2012 edition)was used to establish a fingerprint,and common peak information was obtained;an external standard method was used to establish a standard curve,and the common peaks of the compound were quantified;a chemometric analysis model was established,including cluster analysis,principal component analysis and orthogonal partial least squares-discriminant analysis,to analyze the differences between different batches,and screen out differential markers;the total sugar and total flavonoid content,hygroscopicity,moisture content and long-term stability of the compound were determined,and a tentative standard for the compound was proposed.The experimental results showed that: The extraction rates of flavonoids from Yantuo and polysaccharides from baicalin were between 21.54%~35.66% and 2.03%~11.88%,respectively;The compound contained 1337 compounds,among which flavonoids,phenolic acids,amino acids and their derivatives,organic acids,lipids and sugars were 306,282,122,117,114,73 species respectively,totaling 13 categories of compounds;The established HPLC conditions used methanol as solvent,0.1% phosphoric acid-water solution as mobile phase for gradient elution;The detection wavelength was 310 nm,and after methodological investigation,its precision was high,stability was strong and repeatability was good,which could be used for subsequent research;The similarity of characteristic fingerprint of compound was between 0.615~0.994,with an average similarity of 0.942;The content ranges of 15 batches of yantuo glucoside,yangmei glucoside,buddleoside A,baicalin and quercetin were 1.24~135.17 mg/g,0.12~7.34 mg/g,0.864~5.83 mg/g,5.30~19.58 mg/g,and 2.94~29.02 mg/g respectively;Cluster analysis and principal component analysis yielded four classes,and two differential markers(VIP>1)were screened out,which were yantuo glucoside and baicalin;One quality control system for flavonoid and baicalin polysaccharide compound was obtained;In the quality standard inspection of compound,the total flavonoid,total sugar content,hygroscopic weight gain rate,moisture content were 10.14%~16.44%,5.94%~40.04% 0.49%~0.70%,9.44%~14.88%and 0.49%~0.70%,respectively,and the quality was stable within 6months;The tentative hygroscopic weight gain rate should not be higher than 2%,and moisture content should not be higher than 15.1%.In summary,the compound of flavonoids and baicalin polysaccharides contained various chemical components,with flavonoids and phenols being the most;The fingerprint similarity was high,five main components were quantitatively determined,and the differential marker components of compound were yantuo glucoside and baicalin;The quality standard of compound was proposed,which could provide theoretical basis for subsequent research and development of Chinese veterinary medicine. |
