The liver,as the most important metabolic organ,plays a crucial role in the regulation of the homeostasis of glucolipid metabolism in the body.In the process of livestock production,along with the widespread promotion of intensive and large-scale farming methods,fatty liver syndrome commonly occurs in livestock and poultry farming.When livestock and poultry become ill,their production performance is seriously reduced and even causes death,which seriously affects the economic benefits of livestock and poultry breeding and poses a great threat to the development of the livestock industry.Tussilagone is a compound extracted from the natural plant Tussilago farfara L,which has a variety of biological functions and is inexpensive and non-toxic,making Tussilagone of some research value in the field of livestock and poultry feed development.In vitro,we investigated the modulatory effect of adding 70μM of Tussilagone on the steatosis of HepG2 hepatocytes using oleoyl palmitate(OAPA)-induced HepG2 cells as a model.In vivo,C57BL/6J male mice were used as animal models and were randomly divided into four groups of seven animals each.(1)standard diet group(NC);(2)high-fat diet group(HFD);(3)high-fat diet plus 15 mg/kg Tussilagone group(TUS-15);(4)high-fat diet plus 15 mg/kg Tussilagone group(TUS-30).The effects of Tussilagone on body weight and body fat mass of mice on high-fat diet,and on changes in glucose tolerance and insulin sensitivity in mice on high-fat diet were observed.To detect the effect of Tussilagone on the changes of antioxidant indexes in the liver of mice on high-fat diet;The effect of Tussilagone on energy metabolism changes in high-fat diet mice was recorded by metabolic cage monitoring system.The results showed that Tussilagone supplementation could reduce the contents of triglyceride and total cholesterol in HepG2 cells induced by OAPA,and reduce the synthesis of lipid droplets.The addition of Tussilagone can improve the glucose uptake capacity of HepG2 cells,and increase the glycogen content,mitochondrial number and ATP content in HepG2 cells.Tussilagone supplementation decreased the antioxidant content in HepG2 cells and increased the activity of antioxidant enzymes in HepG2 cells.The body weight of mice in the high-fat diet group was significantly increased,and the weight gain induced by high-fat mice was significantly decreased after Tussilagone treatment.NMR and CT analysis showed that Tussilagone treatment reduced the fat content and lipid accumulation in the liver of high-fat diet mice.Tussilagone treatment also improved dyslipidemia,glucose tolerance and insulin sensitivity of high-fat diet mice.Tussilagone treatment significantly increased the activity of antioxidant enzymes in liver of high-fat diet mice.After Tussilagone treatment,the diurnal O2consumption,CO2production and heat production of high-fat diet mice were significantly increased,and the energy metabolism was increased.In conclusion,the results of in vivo and in vitro experiments show that Tussilagone can improve lipid-induced hepatic steatosis by improving energy metabolism and antioxidant activity,it provides a theoretical basis for the rational development and research of Tussilagone as a feed additive for livestock and poultry.
