As a global epidemic parasitic disease,chicken coccidiosis is a protozoal disease caused by a variety of coccidia such as Eimeria tenella,which easily causes morbidity and mortality in chickens.At present,chemical drugs are the main way to control coccidiosis epidemic.However,due to the long-term abuse of chemical drugs,it is easy to lead to the increase of drug residue and drug resistance in the body,Previous studies have found that Chinese herbal medicine has the effect of killing insects and inhibiting insects,and because of its advantages of small toxic side effects,low rate of drug residues,and difficulty in producing drug resistance,so the use of Chinese herbal medicine or Chinese herbal medicine extracts or preparations to prevent coccidiosis has become a popular research direction nowadays.As a new type of Class II veterinary drug preparation,Boluohui San is a natural plant-derived pharmaceutical feed additive,100% pure plant extraction,which can be used as a feed additive for a long time(it has been used on a large scale in the EU since2000),and there is no withdrawal period.The main active alkaloids such as sanguinarine,chelerythrine and allocryptopine contained in Boluohui San have been confirmed to have significant effects on animal organisms such as "bowel reduction,anti-inflammatory and growth promotion".This topic is the first to study the effect of Boluohui San on coccidiosis chicken,which was studied from the following two aspects: 1.The therapeutic effect of Boluohui San on Eimeria tenella,2.The preventive effect of Boluohui San on Eimeria tenella.First,a chicken coccidial infection model was established,then administered,and finally the effect of Boluohui San on Eimeria tenella was comprehensively explored by calculating the anticoccidial index,measuring some biochemical parameters in serum,and detecting intestinal tight junction proteins and inflammatory cytokines.According to the therapeutic test in this paper,the two dose groups of 1000mg/kg and2000mg/kg were set up with the dose of 300-500mg/kg of normal feed additives as a reference,and the anticoccidial indices of the two groups were 145.6 and 145,respectively,which were consistent with the internationally recognized standards for low-potency anticoccidial drugs.However,due to individual differences in chickens,the survival rate of each group after infection with coccidia was 100%,which had a greater impact on the actual results,and the bloody stool was not improved,so it was only used as a reference.Infection with coccidia generally leads to a decrease in the contents of TP,GLB and TG in serum and an increase in the contents of ALT and AST.However,the results of this therapeutic trial showed that there was no significant difference in increasing the contents of TP,GLB and TG in the blood and decreasing the contents of ALT and AST.The results of relative expression of ZO-1 and Occludin genes showed that Boluohui San had a certain effect of protecting the intestinal barrier;the results of relative expression of TNF-α and IL-6 genes showed that for intestinal inflammation caused by coccidia,the therapeutic effect of Boluohui San was not significant.Therefore,it is concluded in this paper that Boluohui San is not effective in the treatment of chicken coccidiosis by adding two doses of 1000 mg/kg and 2000 mg/kg to the diet.According to the results of the prevention test in this paper,Boluohui San scattered chicks were fed 5 days before infection with chicken coccidia and divided into four groups:200 mg/kg,400 mg/kg,800 mg/kg and 1600 mg/kg.The anticoccidial index of each group was 144.7,151.8,162.7 and 157.7,respectively,which met the standard of low-efficiency anticoccidial drugs,and even the efficacy of 800 mg/kg group was evaluated as moderate anticoccidial effect.Since the survival rate of chickens in each group was 100%,which had an effect on the actual results,it was only used as a reference.The biochemical parameters were detected,and it was found that the groups of Boluohui San had a weak increasing effect on TP,GLB and TG in the serum decreased by coccidial infection,and also had a certain increasing effect on the decrease of ALT and AST in the serum;the results of the relative expression levels of ZO-1 and occludin genes showed that Boluohui San had a certain protective effect on the intestinal barrier;the results of relative expression of TNF-αand IL-6 genes showed that Boluohui San had no significant preventive effect on intestinal inflammation triggered by coccidia.It is concluded that 200 mg/kg,400 mg/kg,800 mg/kg and 1600 mg/kg of Boluohui San have little effect in preventing chicken coccidiosis,and800 mg/kg is superior to the other Boluohui San groups.The above test results showed that although Boluohui San was affected by many conditions under the test conditions,it had a weak effect in reducing the discharge of some coccidial oocysts,and the preventive effect was very well compared with the therapeutic effect for the influence of other indicators in the body.Therefore,in clinical use,according to the compatibility of drugs,we can find a new insecticidal effect of traditional Chinese medicine(such as areca nut leaves)and Boluohui San to act together,configure premix,so as to achieve better protection of the intestine and anticoccidial effect.
