| Germplasm resources of colored potato were lacking in China,and there are few varieties that have been developed and widely promoted in production.It is urgent to cultivate new varieties of colored potato with high quality,high yield and strong adaptability.In this study,six new colored potato lines NNCS-01,NNCS-05,NNCS-06,NNCS-07,NNCS-33,and NNCS-37,which were hybridized in our research group in the early stage,as materials.The MIN-021 introduced from abroad was used as the control.phenotypic traits,pollen fertility,chromosome behavior of pollen mother cell meiosis metaphase Ⅰ(PMCM Ⅰ)and genetic differences at DNA molecular level were analyzed using conventional methods such as field planting observation,chromosome preparation,and SSR molecular marker technology.The main results are as follows:1.The main phenotypic characteristics and growth stages of above-ground parts of six new colored potato strains were identified,including plant height,stem diameter,number of main stems,number of branches,flower color,leaf color,stem color,and plant type.The new strains NCNS-07 and NCNS-37 were medium-early maturing,with a growth period of 85 to 90 days;The new strains NCNS-01,NCNS-05,NCNS-06 and NCNS-33 were all medium ripe with a growth period of 95 to 105 days.2.Six new strains have concentrated tuber forming ability,shallow tuber bud eye,showed significant differences in potato shape,skin and flesh color,and skin smoothness.The number of potato producing per plant ranged from 5.83 to 9.50,and strain NCNS-06 had the highest and strain NCNS-07 had the lowest.The yield per plant of each strain ranges from 816.50 to 1102.40 g,and the rate of commercial potato ranges from 77.64 % to 96.17 %.The yield per plant of NNCS-01 strain was the highest,and the rate of commercial potato number of NNCS-33 strain was the highest.3.The contents of dry matter,starch and protein of the six new strains were in the range of 16.23 % to 22.89 %,11.90 % to 15.99 % and 1.71 % to 2.98 %,respectively.The NNCS-01 strain shows the highest performance and could be used for whole powder processing.The reducing sugar content of each strains was low,ranging from0.018 % to 0.043 %.The content of vitamin C was between 14.41 and 21.38mg/100g·FW,with NNCS-06 strain being the highest;The anthocyanin content ranges from 141.32 and 572.39 mg/kg·FW,with NNCS-37 strain being the highest,which was a new strain with high anthocyanin content.4.The pollen fertility rates of six new colored potato strains NNCS-01,NNCS-05,NNCS-06,NNCS-07,NNCS-33 and NNCS-37 were 44.00 %,66.78 %,74.44 %,60.70 %,78.10 % and 80.22 %,respectively.Their chromosome pairing configurations were 8.45Ⅰ+6.09Ⅱ+7.35Ⅲ+1.33Ⅳ、6.91Ⅰ+7.13Ⅱ+5.73Ⅲ+2.41Ⅳ、5.19Ⅰ+9.16Ⅱ+4.51Ⅲ+2.74Ⅳ、6.55Ⅰ+8.35Ⅱ+4.97Ⅲ+2.46Ⅳ、4.02Ⅰ+10.15Ⅱ+3.40Ⅲ+3.37Ⅳ and 3.31 Ⅰ+10.42 Ⅱ +2.99Ⅲ +3.72 Ⅳ,showed that there were some differences among all strains in cytogenetic level.5.A total of 151 polymorphic band sites were obtained by PCR amplification of genomic DNA of six new color potato strains and control materials using 10 pairs of SSR suitable primers,and the ratio of polymorphic sites was 86.78 %.Using specific SSR primer C42,an SSR fingerprint map was established which could clearly identify six new colored potato strains and control materials.Based on the mean genetic distance(GD value)of 0.62,the seven materials were divided into three categories:new strains NCNS-37,NCNS-06,NCNS-07,NCNS-05 and control MIN-021.The new strains NCNS-33 and NCNS-01 each belong to a separate category. |
