| The morphology of panicle is one of the most important factors to affecting yield and it has a great and practical significance on study the functions of panicle development-related g enes in rice.Zhang Dabing’s team which comes from Shanghai Jiaotong University screened a group of genes related to rice panicle development through gene chip technology.In this thesis,we selected one of its functions---unknown functions genes OsQ828 to do the research,the main results as following:(1)Bioinformatics analysis:we used some bioinformatics analysis soft wares and websites to predict the function of OsQ828,we found OsQ828 is on the chromosome 3,and the genomic sequence length is 1,002 bp,the CDS length 609 bp.The prediction of protein phosphorylation indicates it has two Serine sites,three Threonine sites,but no Tyrosine sites.The prediction of transmembrane suggested it’s a non-trans membrane protein.The 1,500 bp promoter sequence element upstream of ATG predicts that there are a large number of light regulatory elements,tissue-specific expression elements,hormone response elements and stress response elements in the promoter sequence.(2)Mutant plant phenotype and histological analysis:in this study,we obtained a mutant plant of OsQ828 by genome-directed editing.The result showed that OsQ828-2has obviously different morphology from the wild type,whose grain size increased significantly and long awn.Observing under the microscope,we found OsQ828-2 has no defects in floral organs,there is no difference between the wild type and OsQ828-2.The scanning electron microscope displayed a great distinction in the volume and arrangement of cell from wild type and OsQ828-2,which showed that the glume cells arrangement of OsQ828-2was not as concentrate as wild type,and cells volume obviously increased,which suggests that it was the increased cells volume that leaded to grain size increasing.(3)Analysis of expression patterns:The results of tissue expression prediction and determination showed that OsQ828 had higher expression levels during the development of leaves and panicles.OsQ828 is localized in the nucleus by using tobacco transient expression system. |
