The global epidemic caused by the new coronavirus in recent years has brought widespread attention to biosafety events caused by pathogenic microorganisms,and also accelerated the research speed for pathogenic microorganism detection.To address the problem of how to achieve rapid detection of pathogenic microorganisms,this paper takes tobacco ringspot virus as the detection object,designs an impedance detection analysis platform for tobacco ringspot virus based on microfluidic impedance method,establishes an equivalent circuit model to analyze the experimental results,determines a better detection frequency for detecting tobacco ringspot virus,and establishes an impedance-concentration regression model based on this frequency for the detection of tobacco ringspot virus by tobacco ringspot virus detection device.Based on this model,the impedance-concentration regression model for tobacco ringspot virus detection device was established;based on this model,a tobacco ringspot virus detection device was designed using the AD5933 impedance detection chip,and a comprehensive test study was conducted on the developed tobacco ringspot virus detection device through various testing methods to verify the feasibility of the tobacco ringspot virus detection device and provide a technical guarantee for the field detection of pathogenic microorganisms.The main findings of this paper are as follows.(1)An impedance-concentration regression model was established for the detection of tobacco ringspot virus by microfluidic impedance method.The detection mechanism of tobacco ringspot virus impedance measurement was studied,and a gold forked finger electrode microfluidic chip and chip fixture were designed to build a detection and analysis platform for tobacco ringspot virus impedance detection.An equivalent circuit model for the detection of tobacco ringspot virus was established to analyze the experimental results,and the optimal detection frequency for the detection of tobacco ringspot virus was determined to be 63.9 Hz.A tobacco ringspot virus concentration-impedance regression model was established at this optimal frequency: Z=125.49log(C)+652.93(R2=0.9939).(2)The hardware module of the tobacco ringspot virus detection device was designed,including microfluidic chip,impedance measurement module,main control module,display module,alarm module and power supply module.The software module,including impedance measurement module,transmission module and display module,was designed.The development of a tobacco ringspot virus detection device was completed,which can quantitatively detect the concentration of tobacco ringspot virus at a better detection frequency.(3)The detection performance of the tobacco ringspot virus detection device was tested comprehensively,and the stability,reliability and specificity of the tobacco ringspot virus detection device were tested,and the testing performance was good.Compared with the traditional tobacco ringspot detection method,the tobacco ringspot virus detection device has the characteristics of low detection limit,short detection time and high specificity.
