| Chinese jujube(Ziziphus jujuba)is a species of Rhamnaceae and is an important economic forest fruit tree in China.At present,Chinese jujube are mainly distributed in the middle and lower reaches of the Yellow River and southern Xinjiang,where soil is arid and salinity,limiting the development of jujube trees.Chinese jujube is a typical mycorrhizal tree species,which is an important mechanism for Chinese jujube to adapt to drought and salinity environment,so it is of great significance to establish an efficient mycorrhizal seedling system.In traditional seedling breeding techniques,Chinese jujube have relatively low reproductive efficiency and long cycle.In the established Chinese jujube tissue culture system,there are still problems such as high technical requirements,high cost,low survival rate of transplanting seedlings and large differences in tissue culture systems between different varieties,Therefore,it is of great significance to optimize the hormone concentration and ratio of Chinese jujube tissue culture seedlings in the culture stage,establish an efficient and stable Chinese jujube rapid proliferation system,and cultivate mycorrhizal seedlings to realize the large-scale propagation and promotion of Chinese jujube planting.In this study,the effects of 6-BA,TDZ,IBA,NAA,etc.with different concentrations and ratios on the proliferation,elongation,rooting and other factors of wild jujube and’Junzao’ propagation and rooting stage were studied.The optimized parameters for the rapid proliferation of wild jujube and ’Junzao’ was obtained.In addition,AMF was inoculated during the refining of wild jujube and ’Junzao’ rooting seedlings,and the effects of AMF on the survival rate,seedling height and growth status of jujube rooting tissue culture seedlings were studied,so as to realize the cultivation of mycorrhizal seedlings.The main findings are as follows:(1)In exploring the effects of different basic medium and TDZ concentrations on the proliferation culture of wild jujube and ’Junzao’,it was found that the best basic medium suitable for the proliferation culture of wild jujube and ’Junzao’ was modified MS.In the modified MS medium,the proliferation coefficients of wild jujube and ’Junzao’ were 4.15 and 3.38,respectively,and the plant growth state was good.In B5 medium,the proliferation efficiency was the lowest and the growth was poor.The TDZ concentration with the best effect on the proliferation and culture of wild jujube and ’Junzao’ was 0.012 mg/L.At this concentration,the proliferation coefficient of wild jujube plants reached 4.35,the plant growth was good and robust,and the leaves were tender and green;the proliferation coefficient of ’Junzao’ reached 4.62,the plant proliferation efficiency was high and the growth was good,and the leaves were large and green.(2)When screening and optimizing the hormone concentration and ratio of wild jujube and ’Junzao’ subculture,it was found that the optimal hormonal concentration combination of wild jujube subculture was 0.8 mg/L 6-BA+0.008 mg/L TDZ,1.0 mg/L 6-BA+0.004 mg/L TDZ,the proliferation coefficient was 3.01~3.80,and the average elongation of stem segment was 2.36~2.46 cm;The optimal hormone concentration combination for subculture of ’Junzao’was 1.2~1.5 mg/L 6-BA+0.2 mg/L IBA,the proliferation coefficient was 3.71~3.73,and the average elongation of stem segments was 2.44~2.57 cm.(3)In the optimization of rooting culture,it was found that the best rooting medium for wild jujube was 1/2MS+0.1 mg/L IBA+0.1 mg/L NAA+0.3 g/L AC,the rooting rate was 84.44%,the average root number was 1.65 piece,the average root length could reach 6.01cm,the root system was long and adventitious roots,and the best rooting medium for ’Junzao’was 1/2MS+0.6 mg/L IBA+0.3g/L AC,and the rooting rate was 82.22%.The average root number was 2.03 piece,the average root length was 1.27 cm,the root system was thick and the aerial part grew well.(4)When exploring the effect of AMF inoculation on the rooting seedlings of wild jujube and ’Junzao’,it was found that after 45 days of inoculation,the survival rate of transplanting seedlings of wild jujube and ’Junzao’ rooting tissue culture seedlings reached 92.59%and 96.30%,respectively,which were 22.75%and 18.56%higher than the control.After 90 days of inoculation,the seedlings of wild jujube and ’Junzao’ were 2.05 and 1.59 times higher than those of the control,respectively.After transplanting for 90 days,the total biomass of wild jujube and ’Junzao’ inoculated with AMF was significantly higher than that of the control,which was 2.11 and 1.47 times that of the control,respectively.From the perspective of growth state,the wild jujube and ’Junzao’ control plants had poor growth status,the leaves were yellow-green and there were few new shoots at the top,while the wild jujube and ’Junzao’ inoculated with AMF had thick green leaves,fast growth rate and continuous new shoots growing at the top.Therefore,AMF inoculation can significantly improve the transplanting survival rate and biomass of seedlings rooted tissue culture seedlings of wild jujube and ’Junzao’,accelerate the growth rate of seedlings and significantly improve the growth status of seedlings. |
