Cryptosporidium and Enterocytozoon bieneusi are two common intestinal protists,they can infect humans and animals,and it is found in most countries.In previous studies,it was found that there were significant differences in intestinal infection of these two pathogens in sheep or goats from different countries,and genetic characteristics were also diverse.In this study,Jianzhou big-eared goat was selected as the research object.Based on molecular biological methods,the infection situation of Cryptosporidium and E.bieneusi in their intestinal tract was detected,and the species(subtype)/genotype distribution was analyzed.(1)Based on the SSU rRNA gene locus and gp60 gene locus of Cryptosporidium,a total of 18 positive samples were amplified.They are C.xiaoi(n = 15),C.parvum(n = 1)and C.ubiquitum(n = 2).C.xiaoi was the dominant species,and the overall infection rate was 2.3%(18/770).The infection rate of Cryptosporidium was the highest in Lezhi county(27.1%,13/48).No positive samples were detected in Hefeng,Jinma and Yunlong town.The infection rate of Cryptosporidium in Jianzhou big-eared goat at different sampling sites was significantly different(P<0.01).Among different age groups,the infection rate of Cryptosporidium was the highest in 4-6 months old big-eared goat(11.5%,16/139),with infection rates of 2.5%(1/40)and 0.2%(1/591)in ≤3 months old and 7-9 months old goat,respectively.Among different sample sources,the infection rates of samples from free-range farmers,large farms and small farms were4.6%(13/284),2.3%(3/133)and 0.6%(2/353),respectively.There was a difference of significance in the infection rates of Cryptosporidium among different sample sources and age ranges(P<0.01).The successfully amplified samples were amplified based on gp60 gene locus,and a gene subtype,XIIa,was identified after BLAST comparison of the amplified samples.The results of this study showed that the infection rate of Cryptosporidium was low in big-eared goat,and Lezhi county is the highest,with C.xiaoi as the dominant species.(2)Based on the internal transcribed spacer(ITS)region of E.bieneusi ribosomal DNA,43 positive samples were amplified,and the overall infection rate was 5.6%(43/770).At different sampling sites,the infection rate of E.bieneusi was the highest in Pingquan town(14.6%,6/41),and the infection rate of E.bieneusi in Yunlong Town and Jinma town was 13.0%(12/92)and 6.9%(7/101),respectively.And the infection rate of E.bieneusi in different regions was significantly different(P<0.01).The infection rates of E.bieneusi were 12.2%(17/139)and 4.4%(26/591)in sheep aged 4-6 months old and 7-9 months old,respectively.No E.bieneusi was detected in goat aged ≤3 months old.In different sampling sources,the highest infection rate of E.bieneusi was 13.5%(18/133)in large goat farms.The infection rates of E.bieneusi in Jianzhou big-eared goat from different sample sources and age groups were extremely significant difference(P<0.01).Five genotypes were found,namely CHG3(n = 37),CHG7(n = 1),CHG11(n = 1),BEB6(n = 1)and CHG28(n = 3),in which CHG3 was the dominant genotype.The results of this study indicated that the infection rate of E.bieneusi was low,and its genotypes were diverse at different sampling sites.The infection rates of Cryptosporidium and E.bieneusi in Jianzhou big-eared goat are low,and the infection rate of lambs is generally higher than that of adult goat.E.bieneusi genotypes are diverse.This study provides data for the investigation of enterogenic pathogens in Jianzhou big-eared goat and its specific investigation with other goats.
