| Hemerocallis citrina Baroni,also known as enoki and forget-me-not,is a perennial herb belonging to the genus Daylily of the family Afokoliaceae,and its English name is "Daylily".In recent years,there are many varieties of daylily in China,but the differences between varieties are not obvious,which is due to long-term repeated hybridization and the traits have become mixed.In order to make a breakthrough in the breeding of daylily,it is necessary to use new germplasm resources,so this experiment analyzes the phenological period,phenotypic traits,quality traits,drought resistance and other agronomic traits of the collected daylily varieties and analyzes the genetic diversity of the daylily germplasm resources through SSR molecular marker technology,so as to fully explore and screen specific varieties,and provide a theoretical basis for improving the breeding timeliness,production utilization and construction of daylily germplasm resource bank.1.There were obvious differences in the phenotypic traits of 25 kinds of daylily,and the coefficient of variation of 19 phenotypic trait pairs was in the range of 6.61% to58.73%,the largest coefficient of variation is the number of tillers,and the smallest coefficient of variation is the diameter of the inflorescence.The correlation analysis found that most of the traits of the 25 varieties of daylily were very significant or significantly correlated,and most of the traits of daylily affected each other.The principal component analysis found that there were 5 components with characteristic values greater than 1,and the cumulative contribution rate reached 77.03%,The comprehensive score can analyze that the top five varieties are Guangling No.2,Licheng No.5,Tonghua No.1,Mengzihua and Jiaocheng No.2.Through Q-clustering analysis,it was found that there were both differences and certain correlations among different varieties of daylily.At a genetic distance of 10,the 25 breeds can be divided into 4 categories.The first category contains 4varieties,including Datong daylily,most of which are wild-type varieties with higher yields;The second type is only Hunan daylily,which has smaller plants and earlier flowering period;The third category contains 7 varieties,mainly Shaanxi daylily,etc.,and the flowering time is relatively similar;The fourth category contains 13 varieties,mainly Henan daylily,which has good monoflower traits and good plant growth.This fully reflects the difference and diversity of phenotypic traits of 25 daylily varieties.2.25 kinds of daylily quality traits also have obvious differences.After experimental research,the varieties with the highest content in the four quality traits of soluble total sugar,reducing sugar,soluble protein and flavonoids were screened,namely: Guangling No.2,Zuoquan No.1,April Flower and Sichuan Quxian endemic species.Principal component analysis found that,it can be seen that there are two components with characteristic values greater than 1,and the two principal components are related to the two quality traits of flavonoids and soluble sugars,respectively.3.The drought tolerance of the 25 varieties of daylily also showed significant differences.April flower,Mengzihua and Licheng No.5 have relatively high ex vivo water loss rates,while Zezhou No.2 and Baihua have relatively low ex vivo water loss rates.Licheng 5 had the highest relative conductivity at 13.30%.White flowers had the lowest relative conductivity at 4.11%.According to the evaluation of drought resistance,Baihua has the best drought resistance and Licheng No.5 has the worst drought resistance.4.There were obvious differences in phenological periods among the 25 varieties of daylily.Early maturing varieties include Hunan daylily and other varieties,and the plants are small.Medium-ripe varieties include varieties such as Datong daylily,and the yield is higher.Late-maturing varieties include Shaanxi Da Li Shayuan hua and other varieties,and most varieties have good single-flower traits.5.In this experiment,a total of 46 pairs of SSR primers were used for PCR amplification reaction,and 20 pairs of primers were screened to effectively amplify daylily.PCR amplification reaction was performed on 65 cultivars of daylily,and the results showed that: A total of 97 bands were amplified from the 20 pairs of primers,of which 85 were polymorphic bands,and the percentage of polymorphisms was 85.98%.In the end,99 allele fragments and 69.084 effective allele fragments were obtained.The PIC value of each primer is greater than 0.5,the 20 primers were highly polymorphic in 65 varieties of daylily.The heterozygosity was greater than the expected heterozygosity,and it was clear that there was high genetic diversity in the germplasm resources of 65 species of daylily.The cluster analysis of 65 pieces of daylily material was carried out,and the genetic distance was about 0.54,and 65 varieties could be divided into 3 categories,Dao jian hua was divided into one category,Xia mian hua、Xia mian bian yi hong hua and Shang ying Shuang ji hua was the second category,and the remaining materials could also be divided into the third category.At a genetic distance of around 0.60,wild daylily varieties in all varieties have been separated It shows that different varieties of daylily began to evolve separately at a very early stage. |
