In the process of pepper seed breeding,there are often problems such as slow germination,low germination rate,and poor uniformity,especially after years of storage,the quality of the seeds decreases significantly.To address these issues,different seed pretreatment techniques are often used in production to improve the vitality of chili seeds,improve the utilization rate of aged seeds,and reduce the waste of seed resources.The main technical methods for seed pretreatment include seed cleaning and grading,seed priming,seed coating treatment,etc.Seed cleaning can distinguish and separate seeds and impurities based on their different physical properties.Seed priming is based on the biological basis of seed germination,using physical or chemical methods to treat seeds,promote seed germination,increase seed germination rate,and ultimately achieve the goal of improving seed germination ability.Seed coating refers to mixing seeds with characteristic,increasing their resistance to stress and germination ability.In this study,in order to explore the optimal pretreatment technology for chili seeds,“Ha Jiao 6”was used as the experimental material,and the cleaning and grading conditions of chili seeds were screened by wind separation grading and salt solution grading,respectively.A total of 8 substances,including plant immune inducers,amino acids,and other plant growth regulators,were used to explore the optimal priming treatment formula through single substance initiation and composite initiation treatment methods.Based on further screened formula,the seed coating agent was prepared and the seed coating experiments were carried out to explore the possibility of the application of the priming substance in the seed coating agent.The results indicate that:(1)Two methods of wind selection and salt solution classification were used to classify chili seeds.The results showed that the seeds obtained by 8m/s wind screening in the wind selection classification were superior to the control group and other treatment groups in terms of seed germination potential,germination rate and germination index.Both TTC staining and TTC quantitative analysis confirmed that the vitality of seeds retained after grading was significantly better than that of eliminated seeds.The seed germination potential,germination rate and seed index of chili pepper seeds after cleaning and grading with 6% Ca Cl2 solution were higher than those of the control group.Both TTC staining and TTC quantitative analysis verified that the seed vitality retained by grading was significantly better than that of eliminated seeds.(2)In the seed priming experiment,the plant immune inducer substance 900mg/L of Weili had the most significant effect on the improvement of seed germination ability.The germination potential,germination rate,germination index,and seed vitality were 5.18%,16.39%,1.106 and 1.72 higher than those of the control group,respectively.The seeds treated with the composite treatment combination of900 mg/L Weili solution,200 mg/L sodium nitrophenol solution and 50 mg/L aspartic acid solution,showed a significant improvement in germination rate and seed vitality.(3)Through the determination of various physiological indicators of the seeds after the optimal priming treatment,it was found that the 900 mg/L Vitili,800 mg/L Atalin,50mg/L Valine,and 80mg/L proline priming treatments could effectively improve the POD activity,SOD activity,CAT activity,soluble protein content,and soluble sugar content in chili seeds.The treatments of 900 mg/L Vitili,800mg/L Atalin,200 mg/L DA-6,200 mg/L Sodium Nitrophenol,50mg/L Valine,50mg/L Aspartic Acid,and 80mg/L Proline could effectively reduce the content of malondialdehyde in pepper seeds after initiation treatment.(4)In the seed coating experiment,the seed coating agents formulated with “200 mg/L sodium dinitrophenol+50 mg/L aspartic acid” and “900 mg/L vitamin D+50 mg/L aspartic acid” showed the most significant improvement in seed vitality,became more suitable for pepper seed coating treatment.All 7 coating agents formulated by screening different substances could effectively inhibit the growth of pepper wilt pathogen.Among them,the seed coating agent prepared with 200 mg/L sodium dinitrophenol,50 mg/L aspartic acid,and the combination of 200 mg/L sodium dinitrophenol and 50mg/L aspartic acid had better inhibitory effects on Pseudomonas syringa.Considering the improvement of seed vitality and bacteriostasis,the seed coating agent formulated with “200 mg/L sodium nitrophenol and 50 mg/L aspartic acid” had the best effect and could be used as the prototype of a multifunctional seed coating agent.
