Wheat(Triticum aestivum L.)is one of the essential cereal crops in the world today,with tillering and grain quality being important agronomic traits.Amino acids participate in key biological processes in plants,such as growth and development,biotic and abiotic stress,and cell death,and AAPs(Amino acid permease)are crucial amino acid transport protein-related genes in plants.Up to now,there is relatively little research on AAPs genes and their related functions in wheat.In this study,we conducted in-depth analyses of the AAPs(Ta AAPs)gene family in the wheat genome using bioinformatics and other techniques,and studied the expression patterns and functions of Ta AAP11 s.The main research results are as follows:There are three alleles of Ta AAP11 s in wheat,namely Ta AAP11-A,Ta AAP11-B,and Ta AAP11-D.Due to the high expression level in the Axillary meristemin wheat,of Ta AAP11 s were chosen as he research object.Amino acid sequence alignment showed that Ta AAP11-A,Ta AAP11-B,and Ta AAP11-D have high homology(>99%).The phylogenetic tree shows that Ta AAP11 s has different degrees of affinity with AAPs proteins in rice,Arabidopsis,and maize.Subcellular localization results show that Ta AAP11 s proteins are located on the cell membrane.Fluorescent quantitative PCR results show that Ta AAP11 s have high expression levels in spikelets,young spikelets at floret primordium stage and young roots at vegetative stage,Among them,Ta AAP11-B has the highest expression level in the shootapical meristems at the single ridge stage.To explore the function of Ta AAP11 s in wheat,we constructed Ta AAP11 s RNA interference expression vector(Ta AAP11s-RNAi)and constitutive overexpression vector(Ta AAP11-B-OE)for wheat genetic transformation.Compared with wild-type Fielder wheat(control),Ta AAP11-B-OE transgenic wheat showed increased tiller number,root length,and grain volume weight,while Ta AAP11s-RNAi transgenic wheat showed reduced tiller number,shortened root length,and significantly decreased grain volume weight.Compared with wild-type Fielder wheat,the neutral and basic amino acid content of Ta AAP11-B-OE transgenic wheat was significantly increased,while Ta AAP11s-RNAi transgenic plants showed the opposite trend.Therefore,we speculated that Ta AAP11-B may affect the distribution of certain neutral and basic amino acids,then regulate the growth and development of wheat tillering,grain and other processes.Our results provide a molecular basis for understanding the function of AAPs family in wheat,and lay a theoretical foundation for yield improvement and character improvement in wheat.
