Inflammatory bowel disease known as ulcerative colitis(UC)affects the mucosa and submucosa of the colon and is characterized by dysbiosis of the intestinal flora,immunological problems,and disruption of the intestinal mucosal barrier.The primary therapeutic medications currently used to treat UC include sulphasalazine salts,5-aminosalicylic acid,corticosteroids,and calcium-regulated phosphatase inhibitors,but their prolonged usage might have substantial adverse effects.Probiotics and DF by themselves have been the subject of much research in recent years as a means of UC prevention and treatment,but the therapeutic benefits have been found to be slow and variable.In this study,Bacillus subtilis and Bifidobacterium animalis culture conditions were optimized,probiotics and cabbage dietary fiber were configured in a composite formulation in conjunction with preliminary laboratory studies,and The therapeutic effect of a compound formulation on dextran sodium sulfate(DSS)-induced acute UC in mice was carried out.The principal outcomes were as follows:1.Optimizing the conditions for Bacillus subtilis culture:OD value and viable bacteria count were utilized as evaluation markers,with LB medium serving as the base medium.The findings demonstrated that 37°C,4%inoculum,starting p H 7.0,190 r/min,and a 20 h incubation period were the ideal culture conditions for Bacillus subtilis.The viable bacterial count reached 5.34×10~8 CFU/m L,and the bacteria thrived successfully in the culture conditions with soybean peptone 12 g/L,soluble starch 13 g/L,and calcium carbonate 5 g/L as the primary mechanisms.2.This optimized the conditions for Bifidobacterium animalis cultivation:The MRS medium was employed as the base medium,and the OD value and viable bacteria count were used as evaluation indices.The findings demonstrated that 33°C,6%inoculum,initial p H 7.75,and a 40 h incubation period were the best culture conditions for Bifidobacterium animalis.The nitrogen source(peptone:beef paste:yeast powder=2:2:1),glucose,sodium acetate,triammonium citrate,and dipotassium hydrogen phosphate)served as the key mechanism for the bacteria’s growth under the culture conditions.4.58×10~8 CFU/m L of live bacteria were present.3.Research on the probiotic-dietary fiber composite preparation’s therapeutic effects on acute UC:C57BL/6 mice were gavaged with 30%DSS for 7 d to form a mouse model of acute UC injury.The mice were then treated with 0.9%saline(DSS model group),mesalazine(PC group),probiotics(YSJ group)and probiotic-dietary fibre complex(FH group)for 7 d.The effects of each subject on UC were evaluated by examining the morphology,colon histology,oxidative stress levels and intestinal permeability of the mice,compared with a blank control group(NC group).The findings demonstrated that The DAI score was 1.14,which was significantly lower than that of the DSS model group at 2.46(P<0.05),and the mice in the FH group showed better weight gain,colon length,and spleen weight than those in the DSS model group(P<0.05).In contrast to the NC group,the mucosal tissue of the colon of the mice in the DSS model group was severely damaged and the structure of the colon was altered.Comparatively to the NC group,the intestinal mucosa of the mice in the DSS model group was significantly destroyed,and the structure disappeared;however,the colonic tissue in the FH group was only marginally affected,the structure was largely unaltered,and there was still some sign of the colonic mucosa.Compared to the DSS model group,which had a score of 8.23,the histology score was much lower at 3.38.The blood levels of MPO and MDA in the FH group were considerably lower than those in the DSS model group,at 0.52 U/g and 18.96 mmol/ml,respectively(P<0.05),and significantly higher than those in SOD and GSH,at 177.44 U/g and551.76 mmol/ml,respectively(P<0.05);The levels of the tight junction proteins ocludin and claudin in the colon of the DSS model group were substantially greater(P<0.055)than those of the NC group.The expression of the tight junction proteins ocludin,claudin-1,and ZO-1 in the colon of DSS animals was considerably lower than that of the NC group(P<0.055),but it was significantly higher in the FH group.In conclusion,the probiotic-dietary fiber complex has a significant therapeutic effect on DSS-induced acute UC mice and provides an experimental basis for the treatment of acute UC.It can effectively reduce the inflammatory response,inhibit oxidative stress,and protect the intestinal barrier function.
