| Glycyrrhiza Polysaccharides(GP)is a water-soluble polysaccharide.As one of the main effective components of glycyrrhiza uralensis,it can not only enhance the body’s immunity,but also enhance the body’s antioxidant and other defense capabilities,thus protecting the body and reducing the damage caused by oxidative stress,and because of its low price,it is suitable for the development and application of animal clinical drugs.Mancozeb(MCZ)as a commonly used fungicide,is widely used in crop cultivation,but its residual time in the soil is long,and it also has teratogenic,carcinogenic and embryo implantation effects on human and animal bodies.In the theory of traditional Chinese medicine,the kidney is closely related to the reproductive system.Therefore,this experiment preliminarily explored the effect of GP on the kidney damage of offspring mice induced by exposure to MCZ for 21 and 45 days during pregnancy,and provided help to reduce the kidney damage of MCZ on animals,and also provided a scientific explanation for the kidney protection effect of glycyrrhiza.In order to investigate the effect of GP on renal damage in offspring mice exposed to MCZ during pregnancy,SPF grade 7-week-old female and male Kunming mice were selected for the experiment.The mice were adaptively raised for one week and then kept in cages.At 8 am,the presence of vaginal obstruction was checked,and those who detected vaginal obstruction were changed to cages and set as 0 days of pregnancy.Randomly divide the 0-day pregnant female mice into 5 groups,namely:blank control group,MCZ control group,low dose GP group,medium dose GP group,and high dose GP group,totaling 5 groups.The blank control group was given 0.1 mL normal saline,the MCZ control group was given 150 mg/kg MCZ in the morning,and the 0.1 mL normal saline in the evening,and the GP low,medium and high dose groups were given 150 mg/kg MCZ in the morning.At night,50,100 and 200 mg/kg of GP were administered respectively.The first batch of offspring were taken at 21d,and the second batch of offspring were taken at 45d to collect the blood and kidney of the offspring.Calculate kidney index;Histopathological changes of kidney were detected by HE staining.Determination of Cr and BUN in serum of classic indexes of kidney injury by creatine oxidase and urease respectively;The activities of antioxidant related factors TAOC,GSH-Px,MDA and SOD in kidney homogenate were detected;Detection of iron death-related factors GPX4,FSP1 and TGF-β1 level in renal tissue homogenate by ELISA;Determination of iron death-related factor GPX4 and oxidation-related factor NOX4 and NF-κB p65 mRNA transcription level in renal tissue by qRT PCR.The results showed that compared with MCZ group,GP could reduce the content of Cr and BUN in serum,and reduce MDA,NOX4 mRNA,NF-κB p65 mRNA levels in renal tissue,increases the renal index,T-AOC,SOD,GSH-PX,GPX4 mRNA,GPX4 protein,and FSP1 protein levels in renal tissue.The changes in related indicators and the dose of glycyrrhizin are not completely dose dependent,and most of them are NMDR.The trend of changes between glycyrrhizin dose groups and mancozeb group is opposite to that between mancozeb and blank group.In conclusion,GP can alleviate the glomerular enlargement and tubular cell abscission of 21 day old weaning rats and 45 day old sexual maturity rats induced by MCZ exposure during pregnancy,improve the antioxidant capacity and tolerance to iron death of offspring rats,thus reducing the impact of kidney damage on the body,The changes of its indicators and the dose of GP are not completely dose-dependent,and most of them are non-monotonic dose-dependent. |
