Taenia solium cysticercosis is an important food-borne zoonotic disease which is highly prevalent in developing countries such as Latin America,Africa,and South and Southeast Asia.In recent years,many studies on cysticercosis have confirmed that cysticercosis and its excretion and secretion products can inhibit the host immune response,induce immune tolerance and avoid immune attack by self-regulation.Serine protease inhibitors(serpin)are a superfamily of proteins with conserved structure.Studies have shown that serpin inhibits endogenous coagulation pathway by inhibiting the activity of serpins,which enables worms to evade the immune rejection mechanism of the host.These results suggest that serpin plays a key role in the early survival and development of parasites.In this study,we selected serpin B6 and serpin 4848 proteins to explore.Firstly,the bioinformatics characteristic was evaluated and according to the genesequence,designed specific primers.Prokaryotic expression and purification of deendotoxin recombinant soluble target protein.The results of SDS-PAGE and Western blot showed that the two recombinant proteins had good reactivity and immunogenicity.Secondly,the fluorescence signals of Ts-serpin B6 and Ts-serpin4848 were observed at the suckers of the head segment of cysticercus tissue by immunofluorescence localization test.Therefore,it is speculated that serpin of cysticercus porcine may be secreted outside the cyst through the sucker,and then exert its function on the host.Finally,the possibility of r Ts-serpins mediating or participating in immune regulation mechanism was explored by in vitro experiment.After the interaction between human peripheral blood neutrophils and r Ts-serpins,the regulatory levels of r Ts-serpins on neutrophils cytokines were analyzed by q PCR,ELISA and flow cytometry.The results showed that r Ts-serpins significantly increased the expressions of pro-inflammatory cytokines IL-6,IFN-γ,IL-1β and TNF-α,while decreased the expressions of anti-inflammatory cytokines IL-4 and IL-10.In summary,the study preliminarily suggests that r Ts-serpins can be used as an effective immunomodulatory molecule to regulate the immune response of neutrophils.
