| Early in development,the adaptive immune system is not fully developed,and the body mainly uses the innate immune system to respond to pathogenic attacks.The process of immune response is extremely energy-intensive,and will affect most physiological metabolic function pathways such as protein metabolism and lipid metabolism.The liver is the main organ for processes such as immunity,energy metabolism,and synthesis.Because it is responsible for producing most of the immune molecules in the circulation and contains a large number of innate immune cells.The expression and transcription levels of dynamic genes in the liver reflect the immune regulation activities of the system,so the liver may have the potential to systematically analyze the innate immune mechanism of sheep and goats.Therefore,in this study,sheep and goats were used as experimental subjects,and 30 liver tissue transcriptome data were compared and analyzed in five periods,namely fetal period,1-month-old,3-month-old,6-month-old and 2-year-old.The specific research results are as follows:Through sequence alignment analysis,2168 sheep innate immunity genes and 2340 goat innate immunity genes were obtained,and a total of 27 innate immunity-related pathways of sheep and goats and the corresponding genes of each pathway were obtained,and the innate immunity genes of sheep and goats were constructed.database.Comparing and analyzing the liver transcriptome data of sheep and goats at different stages,differentially expressed innate immune genes were obtained through matching,sheep fetal period and 1-month-old is 109,1-month-old and 3-month-old is 71,3-month-old and 6-month-old is 94,6-month-old and 2-year-old is 53.Goat fetal period and1-month-old is 108,1-month-old and 3-month-old is 47,3-month-old and 6-month-old is63,6-month-old and 2-year-old is 38.Through GO function enrichment and KEGG pathway enrichment analysis,it was found that sheep and goats were mainly enriched in Toll-like receptor signaling pathway,innate immune response,NF-kappa B signaling pathway and TGF-beta signaling pathway during fetal period and 1-month-old regulation.It is suggested that differences in these biological processes and signaling pathways in sheep and goat livers may lead to differences in innate immune function in early development.Through the above analysis,the sheep innate immunity candidate gene BAC5 and the goat innate immunity candidate gene TLR7 were finally screened.Comparative analysis was carried out between sheep and goats in five developmental stages,and the differentially expressed innate immune genes were obtained through screening and matching,including 554 in fetal period,396 in 1-month-old,673 in3-month-old,365 in 6-month-old,391 in 2-year-old.Through stem,GO and KEGG analysis,the genes of sheep and goat fetal stage are mainly related to innate immunity.The other four developmental stages are associated with biological processes such as metabolic pathway,cellular processes,etc.Through WGCNA analysis,five co-expression modules were obtained,each module and its corresponding genes: Blue-4154,Brown-1408,Cyan-867,Grey-277 and Tan-147.After GO and KEGG analysis of the genes in each module,the results showed that the genes in the Blue module were enriched in the pathways directly or indirectly related to innate immunity,TLR2,TLR3,TLR4,TLR6 and TLR7 were obtained in sheep and goat livers.These genes may be important genes involved in the early innate immunity of sheep and goats.The genes of Brown module,Cyan module and Grey module were enriched in pathways related to cell and metabolism,and the genes of Tan module were enriched in disease pathways.Finally,candidate genes TLR2,TLR3,TLR4,TLR6 and TLR7 for early innate immunity of sheep and goats were screened.We can screen the early innate immune candidate genes BAC5,TLR2,TLR3,TLR4,TLR6 and TLR7,effective use of biological technology,combined with innate immune genes and molecular marker-assisted selection,research and develop BAC5,TLR2,TLR3,TLR4,TLR6 and TLR7 agonists for sheep and goats,agonists can be used as inducing adjuvants to provide signals to induce effective innate immune responses so that sheep and goats can produce immune protective effects. |
