| Sclerotinia sclerotinia is one of the main diseases in sunflower production at present.It is an economical and effective method to control sclerotinia sclerotinia by breeding resistant varieties,and it is particularly important to identify and screen germplasm germplasm resistance as a link of cross breeding and study the mechanism of resistance.Due to the influence of uncontrollable environmental conditions,the identification results of traditional field disease nursery identification or field inoculation identification can not reflect the real resistance level.In addition,field identification can not ensure that the identified materials are timely incorporated into the breeding program of the same period,which will affect the breeding process and breeding efficiency.Therefore,a rapid batch identification method is needed to remedy the above defects.Based on this,the hydroponic inoculation method was firstly adopted in this study,and the resistant materials and susceptible materials were qualitatively analyzed by calculating the incidence rate.And on this basis further confrontational material potted the inoculation,the reduced glutathione,glutamate oxaloacetate transaminase,glutamic pyruvic transaminase,beta 1,3 glucanase,chitinase,galacturonic acid polymer enzyme,lignin,oxalic acid,such as eight biochemical indexes were measured,using principal component analysis of comprehensive evaluation The germplasm resources of antibacterial nucleosis were screened out by quantitative resistance classification of the selected resistant materials,and the screening efficiency was improved,which is of great significance for efficient cross breeding of antibacterial nucleosis.The main results of this study are as follows:1.Twenty-two resistant materials WM01,WM02,WM03 and 17 susceptible materials WM06,WM08,WM10 were obtained by hydroponic qualitative analysis.2.The resistant materials were qualitatively analyzed in hydroponic culture,and 4high resistant materials of WM04,WM30,WM31 and WM37,12 resistant materials of WM01,WM02 and WM03,and 6 medium resistant materials of WM07,WM13,WM16,WM19,WM23 and WM28 were obtained by pot method.3.The contents of GSH,GOT activity,GPT activity,β-1,3-glucanase activity,chitinase activity and lignin content in leaves of different sunflower resistant materials were increased in different degrees after inoculation with SCLerotinia sclerotinia.The GSH content of high resistant material was lower than that of medium resistant material before inoculation,but the GSH content of high resistant material was higher than that of medium resistant material after inoculation.The activities of GOT,GPT,β-1,3-glucanase,chitinase and lignin content in leaves of high resistance materials remained at a high level before and after inoculation,while the activities of GOT,GPT,β-1,3-glucanase,chitinase and lignin content in leaves of medium resistance materials were relatively low.These results indicated that these biochemical indexes played an active role in the resistance to sclerotinia infection after inoculation with sclerotinia sclerotinia.The oxalic acid content and PG activity of high and medium resistance materials were increased after inoculation,and medium resistance materials were higher than high resistance materials,indicating that the resistance of sunflower to sclerotinia sclerotinia was negatively correlated with physiological indicators. |
