Animal fatty liver is a common disorder of nutrient metabolism in livestock production,mainly in laying hens and dairy cows,but also from time to time in broilers,laying ducks,fish,sheep and other animals.When livestock and poultry suffer from diseases,the body’s normal metabolism is influenced and production performance decreases,which can even cause livestock and poultry to die,bringing serious economic losses to livestock production.With a wide range and low toxicity,natural small molecules of plant origin have great potential for development as plant-based feed additives.Therefore,we screened 243 natural small molecules using an oleic acid palmitic acid induced HepG2 cell fat accumulation model and identified a natural alkaloid extracted from Radix Isatidis,epigoitrin,which has better lipid-lowering effects.In this study,using HepG2 cells and LMH cells as in vitro models and C57BL/6J mice as in vivo models,we investigated the regulatory effects of Radix Isatidis extract epigoitrin on liver lipid and glucose metabolism induced by high-fat.In vitro experiments were divided into oleic acid palmitic acid control group and a treatment group with the addition of epigoitrin.The results showed that epigoitrin significantly reduced hepatocyte triglyceride(P<0.05)and cholesterol content(P<0.01),enhanced short-chain acyl Co A dehydrogenase activity(P<0.001),promoted glucose uptake(P<0.01)and glycogen storage(P<0.01),increased ATP production(P<0.01),and improved the antioxidant capacity of hepatocytes.The 42 seven-week-old male C57BL/6J mice were randomly divided into 6 groups(n=7)for a 12-week in vivo experiment: normal chow(NC),normal chow + 50 mg/kg epigoitrin(NC50),normal chow + 100mg/kg epigoitrin(NC100),high-fat diet(HFD),high-fat diet + 50 mg/kg epigoitrin(HFD50)and high-fat diet + 100 mg/kg epigoitrin(HFD100)groups.The results showed that epigoitrin significantly inhibited weight gain(P<0.01)and fat accumulation(P<0.001),reduced liver triglyceride(P<0.001)as well as cholesterol content(P<0.05)and alleviated liver steatosis,increased glucose tolerance(P<0.001)and improved insulin sensitivity(P<0.01),and raised energy expenditure(P<0.01)in mice induced by high-fat diet.The results of both in vitro and in vivo experiments indicated that epigoitrin inhibits high-fat induced lipid deposition and alleviates disorders of liver lipid and glucose metabolism,which provides a theoretical basis for epigoitrin to prevent and treat fatty liver in animals.
