Trametes versicolor(L.) Lloyd is also known as Coriolus versicolor,Melaleuca,Baizhi,etc.It has medicinal effects such as nourishing and protecting liver,anti-cancer and anti-cancer,and enhancing human immunity.Related health products and medicines have appeared on the market.Trametes versicolor polysaeeharide and polysaccharidiopeptide are the main pharmacological active ingredients of Trametes versicolor.According to the regulations on page 60 of the 2020 edition of the Chinese Pharmacopoeia,Coriolus versicolor polysaeeharide and polysaccharidiopeptide.At present,the continuous emergence of Trametes versicolor series of medicines and health care products has further increased the demand for Trametes versicolor at home and abroad.Almost all Trametes versicolor used on the market is picked from the wild,and the output cannot meet its production needs.The resources of wild Trametes versicolor are vague,and the resources of Trametes versicolor varieties that can be used for artificial cultivation are not clear,and people’s research on Trametes versicolor mainly focuses on its active ingredients and pharmacological effects.There is a lack of domestication and cultivation of Trametes versicolor and the improvement of polysaccharide content.the study.In this study,the collected 10 strains of Trametes versicolor strains were isolated and identified,and the mother seed culture medium and cultivation material formula of Trametes versicolor were optimized to explore the effects of different carbon and nitrogen sources in the mother seed culture on the mycelium of Trametes versicolor mycelium.The effects of growth speed,density,and biomass were compared.The effects of different cultivation material formulations on the quality,yield,polysaccharide content and extract content of Trametes versicolor fruiting bodies were compared,and 1 to 2 excellent strains were screened.The main findings of this paper are as follows:1.Use antagonism experiment and ITS molecular identification methods to identify the strains of the collected resources of 10 Trametes versicolor varieties.The identification results show that the 10 strains are all Trametes versicolor strains,and the genetic distance between the strains is between 0-0.011.The relationship is very close.The strains YUN2 and YUN4,DB4 and JL2,and YUN1 and HN1 are closely related.2.Six different carbon sources and five different nitrogen sources,as well as different pH values ??were used to screen,and it was determined that the suitable carbon source for Trametes versicolor growth was glucose,the suitable nitrogen source was yeast powder,and the carbon to nitrogen ratio was 15:1.The pH range is 6-7.Then use Box-Behnken experimental design and response surface analysis to carry out a three-factor three-level response surface experimental design,optimize the medium,and the optimized medium dosages are: glucose 21.13g/L,yeast powder 4.94g/ L,pH 6.44,and the best predicted value is 1.874g/100 mL.After further experimental verification,the average dry weight of the measured mycelium is 1.876 g/100 mL of HN1,dry weight of YUN1 1.872 g/100 mL,The dry weight of DB4 is 1.868 g/100 mL,the dry weight of DB5 is 1.862 g/100 mL,the dry weight of DB8 is 1.859 g/100 mL,the dry weight of JL3 is 1.831 g/100 mL,the dry weight of YUN2 is 1.795 g/100 mL,and the dry weight of YUN5 is 1.521 g/100 mL.3.Using sawdust,eucommia,cottonseed husks,wheat bran,etc.as cultivation materials,8different cultivation material formulations were designed to domesticate and cultivate Trametes versicolor.The results showed that Trametes versicolor had a higher yield and Trametes versicolor polysaccharides in the cultivation formula C2(Eucommia 60%,cotton seed hull 20%,wheat bran 17%,sucrose 1%,lime 1%,gypsum 1%)And the extract content is higher than the amount specified in the "Chinese Pharmacopoeia".The cultivation results showed that the highest yield among the 7 strains was strain HN1 with a yield of 1577.8 g,and the lowest yield was strain DB5 with a yield of 1221.5 g.Among them,the highest polysaccharide content is DB4,the polysaccharide content is 6.4%,and the lowest polysaccharide content is strain JL3 with a content of 2.7%;the highest extract content is strain DB4,with an extract content of 24.2%,and the lowest extract content is Strain DB8,the content is 15.9%.Among the 7 strains,the strain YUN2 with the highest water content is 66.46%,and the water content of the 7 strains is between 50%-65%.The fruit body size is between 6-8cm*3-5cm,and the thickness is also between 0.2-0.4cm.According to the test results,combined with the fruit body yield,polysaccharide and extract content,two high-quality Trametes versicolor strains,DB4 and HN1,were successfully obtained.
