| Curcumin and its derivatives are important functional components of curcuma,which have the functions of promoting digestion and absorption,anti-bacterial and anti-inflammatory,lowering lipid and so on.In this study,the effects of curcumin and bisdemethoxycurcumin on abdominal fat deposition of yellow-feathered broilers were studied,and the mechanism of their effects was explored from the Angle of fat metabolism.A total of 420 28-day-old,medium-speed yellow feather broiler hens of similar size were selected.They were randomly divided into 7 groups according to their body weights,with 6replicates per treatment and 10 chickens per replicate.They were fed with curcumin at 0,50,100,and 200 mg/kg and bisdemethoxycurcumin at 50,100,and 200 mg/kg,respectively.Research indicates:1.The aspect of production performance:The supplementation of 100mg/kg and200mg/kg curcumin significantly decreased the feed intake at 29-70 days old(P<0.05).But it had no significant effect on daily gain and feed/gain.2.The aspect of abdominal fat deposition:Adding 50mg/kg,100mg/kg and 200mg/kg curcumin significantly decreased the abdominal fat percentage of yellow-feathered broilers at 70 days old(P<0.05),supplemented with 200mg/kg bisdemethoxycurcumin significantly decreased the abdominal fat percentage of yellow-feathered broilers at 70 days old(P<0.05).3.The aspect of energy efficiency:Both curcumin and bisdemethoxycurcumin improved energy efficiency,which is shown as follows:Adding 100mg/kg and 200mg/kg curcumin significantly significantly increased the energy efficiency of yellow-feathered broilers at29-42 days old,43-56 days old and 29-70 days old(P<0.05).The effects of bisdemethoxycurcumin on the energy efficiency of yellow-feathered broilers showed the same trend,Specifically,the supplementation of 100mg/kg bisdemethoxycurcumin significantly increased the energy utilization rate of yellow-feathered broilers during the three stages of 29-42 days old,43-56 days old and 29-70 days old(P<0.05).Other supplemental levels only increased the energy use efficiency of yellow-feathered broilers during the 43-56 days old period(P<0.05).Effects on blood lipid: Curcumin and bisdemethoxycurcumin decreased serum TC and TG contents of medium speed yellow-feathered broilers,specifically as follows: dietary supplementation of 100mg/kg curcumin significantly decreased serum TC content of yellow-feathered broilers from 57 days to 70 days(P<0.05),adding 50mg/kg curcumin significantly decreased serum LDL-C content of broilers(P<0.05),while bisdemethoxycurcumin only decreased the serum TC content of broilers at 43-56 d regardless of the supplemental dose(P<0.05).Effects on TC and TG in liver: Curcumin and bisdemethoxycurcumin increased the content of TC and TG in liver,and the specific manifestations were as follows : adding200mg/kg bisdemethoxycurcumin significantly increased liver TG content of yellow-feathered broilers at 29-42 days old and 57-70 days old(P<0.05),and significantly increased the liver TC content of 29-42 days old yellow-feathered broilers(P<0.05).Adding100mg/kg bisdemethoxycurcumin significantly increased the liver TC content of 43-56 days old yellow-feathered broilers(P<0.05),the supplementation of 200mg/kg bisdemethoxycurcumin significantly increased the liver TC content of 57-70 days old dyellow-feathered broilers(P<0.05).4.In the aspect of intestinal morphology,curcumin and dimethoxycurcumin improved the intestinal morphology of duodenum of medium speed yellow-feathered broilers,but had no significant effects on jejunum and ileum,which were shown as follows:supplementation of 100mg/kg curcumin significantly increased the villus height and crypt depth ratio of duodenum of broilers at 29-42 days old and 43-56 days old,and significantly decreased duodenal crypt depth at 43-56 days old(P<0.05).Adding 100mg/kg bisdemethoxycurcumin significantly increased the villus height and crypt depth ratio of duodenum in 29-42 days old(P<0.05),the supplementation of 200mg/kg bisdemethoxycurcumin significantly increased the villus height and the villus height and crypt depth ratio of duodenum at 57-70 days old(P<0.05).5.In terms of long chain fatty acids in breast muscle,The supplementation of curcumin and bisdemethoxycurcumin in the diet of medium speed yellow-feathered broilers could decrease the SFA content in breast muscle,and increase the MUFA and PUFA content in breast muscle.which were shown as follows:adding 50mg/kg curcumin significantly decreased the content of tetradecenoic acid in breast muscle of yellow-feathered broilers(P<0.05).Adding 100mg/kg curcumin significantly decreased the contents of Caprylic acid,tetradecenoic acid,eicosadienoic acid,methyl tetracosyl carbonate and docosahexaenoic acid in breast muscle of yellow-feathered broilers(P<0.05),and significantly increased the content of linolenic acid in breast muscle of medium speed yellow-feathered broilers(P<0.05).Adding 200mg/kg curcumin significantly increased the contents of palmitoleic acid,oleic acid,linoleic acid and linolenic acid in breast muscle of yellow-feathered broilers(P<0.05),and significantly decreased the contents of caprylic acid,tridecylic acid,tetradecanoic acid,heptadecanoic acid,stearic acid,eicosadienoic acid,tetoscarbonic acid and docosahexaenoic acid in breast muscle of yellow-feather broiler(P<0.05).Adding 100mg/kg and 200mg/kg curcumin significantly decreased the content of SFA in breast muscle of broilers(P<0.05),and significantly increased the contents of MUFA and PUFA in breast muscle of broilers(P<0.05).Adding 50mg/kg and 100mg/kg bisdemethoxycurcumin significantly decreased the content of heptadecanoic acid in breast muscle of yellow-feathered broilers(P<0.05).Supplementation of 100mg/kg bisdemethoxycurcumin significantly increased the content of linoleic acid in breast muscle of yellow-feathered broilers(P<0.05).Adding 200mg/kg bisdemethoxycurcumin significantly decreased the contents of Caprylic acid,tridecylic acid,Tetradecenoic acid and heptadecanoic acid in breast muscle of yellow-feathered broilers(P<0.05),and significantly increased the contents of linoleic acid,linolenic acid and PUFA in breast muscle of yellow-feathered broilers(P<0.05).6.Liver fat synthesis related indicators:(1)It can reduce the m RNA expressions of liver Fas and ACC,reduce liver fat synthesis,increase the enzyme activity of HSL,and thus promote fat decomposition.Specific manifestations are as follows:adding 200mg/kg curcumin significantly increased liver FAS content of yellow-feathered broilers(P<0.05).The supplementation of 50mg/kg,100mg/kg and 200mg/kg curcumin could significantly increase HSL content in liver of yellow-feathered broilers(P<0.05).The supplementation of50mg/kg,100mg/kg and 200mg/kg bisdemethoxycurcumin significantly increased the liver FAS content of yellow-feathered broilers(P<0.05).Adding 50mg/kg bisdemethoxycurcumin significantly increased the liver ACC content of yellow-feathered broilers(P<0.05).Adding50mg/kg and 100mg/kg bisdemethoxycurcumin could significantly increase the liver HSL content of yellow-feathered broilers(P<0.01).(2)The supplementation of curcumin and bisdemethoxycurcumin in the diet of medium speed yellow-feathered broilers can decrease the m RNA expression of the enzymes related to fat synthesis and affect the β-oxidation of fatty acids.specifically as follows:Adding 200mg/kg curcumin significantly decreased the m RNA expression of ACC in liver of yellow-feathered broilers(P<0.05).The supplementation of 50mg/kg,100mg/kg and 200mg/kg bisdemethoxycurcumin significantly decreased the m RNA expression levels of LPL,ACC and FAS in liver of yellow-feathered broilers(P<0.01).Compared with the control group,the supplementation of 50mg/kg bisdemethoxycurcumin significantly increased the m RNA expression level of CPT1α in liver of yellow-feathered broilers(P<0.01).In conclusion,this study shows that :(1)Curcumin can reduce abdominal fat deposition of medium speed yellow-feathered broilers,and the mechanism of its action is related to the improvement of energy utilization,the reduction of blood lipid,the promotion of fat decomposition and the morphology of duodenum,and the above effects are greater in the late growth period(43-70 days old)than in the early growth period(29-42 days old).(2)The effect of curcumin on fat metabolism is shown as follows: it can increase the enzyme activity of HSL,reduce the m RNA expression of Fas and ACC,and promote the decomposition of liver fat.(3)200 mg/kg curcumin and bisdemethoxycurcumin had the best effect on reducing abdominal fat.(4)Compared with curcumin,bisdemethoxycurcumin has the same effect,and even has better effect on the absorption and utilization of duogene expression of liver FAS m RNA. |
