In recent years,with the improvement of living standards,people’s dietary structure has changed,which has brought a variety of diseases related to metabolism,such as obesity,diabetes,hypertension,enteritis and so on.In animal production,excessive fat deposition will also have adverse effects on animals,such as increased intestinal permeability,decreased muscle-fat ratio,and reduced reproductive performance.The purpose of this study was to investigate the effects of Lactobacillus denderi supplementation in high-energy diet on blood glucose,blood lipid,intestinal morphology,cytokines,inflammation-related protein and gene expression,and intestinal microbial structure in SD rats.In addition,to explore the alleviating effect of Lactobacillus denderi on intestinal inflammation in SD rats under high energy diet,and to provide scientific basis for the development and utilization of Lactobacillus denderi in animal production.A total of 20 healthy male SD rats aged 4 W with similar body weight were selected.They were randomly divided into control group(high energy asepsis)and experimental group(high energy bacteria addition)with 5 replicates per group and 2 rats per replicate.Prefeed for 1W and feed for 11 W.The day before the end of the experiment,the rats were fasted at night,and the blood glucose and lipids were measured by anesthesia on the second day.After the death,the intestinal tract,liver and other tissues were stripped,jejunum and ileum were separated and selected,and the intestinal histological score,cytokines,gene expression of tight junction proteins and inflammatory proteins in intestinal tissue were determined,as well as 16 s RNA sequencing.The test results are as follows:1.Compared with the control group,the supplementation of 0.1% Lactobacillus deli in the diet of experimental groups could reduce the blood glucose and lipid levels,significantly decreased the serum LDL level(P < 0.05),and decreased the levels of BG,TG,HDL and TC,but the differences were not significant(P > 0.05).2.Compared with the control group,the addition of 0.1% Lactobacillus denderi in the diet of experimental group could enhance intestinal compactness and integrity,and alleviate the intestinal barrier function impairment induced by high-energy diet.Among them,the gene expression of intestinal tight junction proteins ZO-1 and Occludin was significantly increased(P < 0.05),intestinal tissue damage score was significantly decreased(P < 0.05),serum DAO and LPS levels were decreased,but the difference was not significant(P >0.05).3.Compared with the control group,the number of lymphocytes and goblet cells in ileum were significantly decreased(P < 0.05),and the expression of proinflammatory factor COX-2 gene in jejunum and ileum was significantly decreased(P < 0.05)in experimental group supplemented with 0.1% Lactobacillus deli.However,there was no significant effect on i NOS gene expression(P>0.05).4.Compared with the control group,the supplementation of 0.1% Lactobacillus demineri significantly increased the gene expression levels of immunoglobulin s Ig A and anti-inflammatory cytokine IL-10(P < 0.05),and significantly decreased the gene expression levels of pro-inflammatory cytokines TNF-α,INF-γ,IL-1β,IL-6 and IL-2(P <0.05).Reduces levels of intestinal inflammation induced by a high-energy diet.5.Compared with the control group,the experimental group supplemented with 0.1%Lactobacillus deli could regulate the structure of intestinal microflora and increase the diversity of intestinal microflora.Reduce the ratio of Firmicutes to Bacteroidetes,Increase the abundance of Lactobacillus,Bifidobacterium,Blautia,Lachnoclostridium,Akkermansia and some butyric acid producing bacteria such as Norankf_Lachnospiraceae,Ruminococcus and other beneficial bacteria.To reduce the abundance of harmful bacteria such as Pseudomonas and Bacillus.Added in the diet with the place on put together is narrated,Germany’s lactobacillus of high-energy diet environment SD rats blood lipid levels,intestinal inflammation levels have reduced effect,this may be due to Germany’s lactobacillus decreased inflammatory factor before the expression of cox-2 and proinflammatory factor,increase the expression of immunoglobulin SIg A and anti-inflammatory factors,at the same time to change the structure of intestinal flora,It increases the diversity of intestinal flora.
