| Objective:This paper mainly studied the effect and toxicity of compound Codonopsis powder on gastrointestinal function of rats and mice,through the effect of compound Codonopsis powder on gastrointestinal function of rats and mice,this paper preliminarily discusses the feasibility of compound Codonopsis powder powder in livestock and poultry breeding to promote animal growth by enhancing gastrointestinal digestion and absorption,and verifies whether compound Codonopsis powder powder has toxic effect through the toxicity test of compound Codonopsis powder powder.The purpose of this study is to provide experimental data and theoretical basis for the safe application of compound Dangshen Powder in livestock and poultry production.Methods: 1.Effect on spleen deficiency mouse model induced by reserpine: 60 ICR mice were randomly divided into 6 groups,half male and half female,with 10 mice in each group.Except for the normal control group,the spleen deficiency model was established in the other groups,and the corresponding drugs were intragastrically administered once a day for 14 days.The body weight was weighed every week,fasted after the last administration,and the blood was taken from the orbit the next day.The levels of gastrin(GAS)and motilin(MTL)in serum were detected by ELISA.The animals were killed and the stomach was dissected for pathological examination and HE staining to observe the changes of gastric mucosa.2.Effect of compound Dangshen Powder on gastrointestinal function of rats and mice:(1)50 SD rats,half male and half female,were randomly divided into 5 groups.The corresponding doses of drugs were given to each group,and the animals in the normal control group were given the same volume of pure water for 7 days.After the last administration,30 min was given intragastric administration of 5 % D-xylose solution 10 m L / kg,and the content of serum xylose in each group was determined;the spleen was dissected,weighed,and the spleen index was calculated.(2)50 ICR mice,half male and half female,were randomly divided into 5 groups.The animals in the normal control group were given intragastric administration of the same volume of pure water for 3 days,and 0.5 m L 0.04 % phenol red solution was intragastrically administered 1 hour after the last administration to determine the intestinal propulsive rate and gastric residual rate of mice.3.Toxicity test:(1)Acute toxicity test: 40 ICR mice,half male and half female,were randomly divided into two groups: experimental group and blank group.According to the maximum dose,the drug was given by oral administration,and the drug was given twice on the same day.Within 4 hours after each administration,the manifestations and characteristics of poisoning,the time of occurrence and recovery of toxic reaction and death were closely observed and recorded for 14 days.The body weight of the animal was recorded every three days.(2)long-term toxicity test: SD rats were given different doses of compound Dangshen Powder by oral administration for 3 months(13 weeks),and the long-term toxicity was observed after 2 weeks of drug withdrawal.80 SD rats,half male and half female,were randomly divided into 4 groups.The experiment was divided into blank control group,low,middle and high dose groups of compound Dangshen Powder.The drug was given by intragastric administration according to the volume of 20 m L/kg for 3 months.Forty rats,half male and half female,were dissected at the end of the drug administration and the end of the recovery period.The examination items included general clinical observation,determination of body weight,food intake,hematology,blood biochemistry,blood coagulation,organ coefficient and histopathological examination.Results: 1.Effect of compound Dangshen powder on spleen deficiency mouse model induced by rehematpine:(1)Body weight: compared with normal control group,the body weight of model control group was significantly decreased after administration(P < 0.05).Compared with the normal control group,there was no significant difference in the body weight of Compound Dangshen Powder low,medium and high groups and Guipi pill groups,indicating that Compound Dangshen Powder could promote the weight growth of spleen deficiency mice.(2)Serum GAS and MTL content: the contents of GAS and MTL in serum were significantly increased in the high-dose group of compound Dangshen powder and Guipiwan group(P < 0.05).(3)Histopathological examination of gastric mucosa: compared with the model control group,the middle-dose group of compound Dangshen powder could improve the gastric mucosa injury.2.Effect of compound Dangshen powder on gastrointestinal function of rats :(1)Compared with normal control group,serum D-xylose in compound Dangshen powder medium-dose and high-dose groups and Gui Pi pill groups was significantly increased(P < 0.01),and spleen coefficient in compound Dangshen powder low-dose,medium-dose and high-dose groups and Gui Pi pill groups was significantly increased(P < 0.01).(2)Compared with the normal control group,the gastric residual rate of compound Dangshen powder high-dose group and Guipi pill group was significantly decreased(P < 0.01),the intestinal propulsive rate of compound Dangshen powder medium-dose group was significantly increased(P < 0.05),and the intestinal propulsive rate of compound Dangshen powder high-dose group and Guipi pill group was significantly increased(P < 0.01).3.Toxicity test: acute toxicity test(1)General clinical observation: no obvious abnormality was observed in autonomic activity,mental state and diet of mice within 0-4 hours and 14 days after oral intragastric administration,and no related toxic reactions and animal death were observed.(2)Body weight: No statistically significant difference was found,indicating that oral administration of Compound Dangshen Powder in ICR mice had no significant effect on body weight gain.No obvious abnormalities were observed on the surface and section of each organ.Long-term toxicity test(1)General clinical observation: During the administration period and the recovery period,compared with the blank control group,there were no abnormalities related to drug toxicity in the appearance,physical signs,behavior and general conditions of the animals in each dose group of compound Dangshen powder.(2)Body weight,food intake,hematological examination,blood biochemical examination,blood coagulation examination,organ coefficient and histopathological examination showed no obvious abnormalities in each dose group of Compound Dangshen Powder compared with the blank control group at the same period.Conclusion: Compound Dangshen Powder can increase the secretion of motility and gastrin,improve the damage of gastric mucosa,improve the absorption rate of small intestine to serum xylose,increase spleen index,accelerate the small intestine propellant rate,and no toxic reaction was observed in compound Dangshen powder. |
