With the complete prohibition of feed antibiotics,it is difficult to protect the health of livestock and poultry caused by pathogenic microorganism infection,stress and other factors is becoming more and more obvious.In order to take precautions against the intestinal infection of livestock and poultry or reduce the occurrence of stress,protect the intestinal health of livestock and poultry,and even the healthy body,and ensure green and healthy ecological breeding.The purpose of this experiment was to explore the preparation technology of Lactobacillus plantarum powder and its application in broiler breeding.Firstly,Lactobacillus plantarum powder was prepared by exploring the fermentation formula and fermentation process parameters of Lactobacillus plantarum and determining the conditions for preparing Lactobacillus plantarum powder,and then the application of Lactobacillus plantarum powder in broiler feeding was studied through animal culture experiment.Experiment.1 Preparation of Lactobacillus plantarum 167 powderDetermination of fermentation formula and conditions:the fermentation time was determined to be 15 h.The fermentation formula was determined as follows:low protein whey powder 32.50 g/L,glucose 30.00 g/L,yeast extract FA25(Angel yeast)2.00 g/L,peptone(Angel yeast)5.00 g/L,magnesium sulfate 0.15 g/L,manganese sulfate 0.03 g/L,dipotassium hydrogen phosphate 2.00 g/L.It is determined that the protective agent ratio is 13%solid powder and 2%silica for powder spraying.Preparation of Lactobacillus plantarum powder:the living bacteria number of the final Lactobacillus plantarum powder was 4.65×108CFU/g.The above results show that the fermentation conditions of Lactobacillus plantarum and the preparation of Lactobacillus plantarum powder are on the premise of controlling the cost,and the best formula is selected according to the number of living bacteria in Lactobacillus plantarum fermentation broth.Experiment.2 Application effect of Lactobacillus plantarum powder in broiler feedingThe purpose of this experiment was to study the application of different doses of Lactobacillus plantarum powder in broiler breeding and analyze its possible mechanism.One hundred and fifty healthy 1-day-old Ross 308 broilers were selected and fed with 0 g/t,500 g/t and 1000 g/t phytobacillus powder in the basal diet with 5 replicates per treatment and 10 broilers per replicate.The experimental period was 28 days.After the experiment,weighing,slaughtering and sampling.The test results show that:(1)Compared with the control group,different levels of Lactobacillus plantarum powder had no significant effect on average daily gain,average daily feed intake and feed gain ratio of broilers at different stages(P<0.05).(2)Compared with the control group,the low-dose Lactobacillus plantarum powder group significantly decreased the content of Ca in plasma of broilers(P<0.01),while the high-dose Lactobacillus plantarum powder group increased the contents of plasma total protein(TP)(P<0.05)and albumin(ALB)(P<0.01).(3)Compared with the control group,the low dose Lactobacillus plantarum powder group decreased the duodenal villus height(VH)and area(P<0.05)and the jejunal crypt depth(CD)(P<0.05).The high dose Lactobacillus plantarum powder group increased the jejunum villus height(P<0.01)and the ratio of villus height to crypt depth(VH/CD)(P<0.05).It decreased the villus height of duodenum(P<0.05)and the depth of jejunal recess(P<0.05).Compared with the control group,the high dose Lactobacillus plantarum powder group upregulated the expression of ZO-1,occludin and claudin-1 genes in the jejunum of broilers(P<0.01),while different levels of Lactobacillus plantarum powder down-regulated the expression of mucin-2 gene in the jejunum of broilers(P<0.01).(4)Compared with the control group,different levels of Lactobacillus plantarum powder decreased the spleen weight of broilers(P<0.05).Compared with the control group,low dose Lactobacillus plantarum powder group upregulated the expression of INF-γand TNF-αin duodenum,TGF-β4 in jejunum,INF-γ,i NOS and TGF-β4 in ileum in broilers(P<0.01).High dose Lactobacillus plantarum powder group upregulated the expression of IL-8,INF-γ,i NOS,TNF-αin duodenum,IL-1β(P<0.05),TNF-αand TGF-β4 in jejunum,i NOS and TGF-β4 in ileum of broilers(P<0.01).(5)Compared with the control group,low dose Lactobacillus plantarum powder group increased the activity of glutathione peroxidase(GSH-Px)and the content of hydrogen peroxide(H2O2)in jejunum of broilers,and increased the content of malondialdehyde(MDA)in ileum of broilers(P<0.01).High dose Lactobacillus plantarum powder increased CAT activity in duodenum(P<0.05),GSH-Px activity and H2O2content in jejunum and MDA content in ileum(P<0.01),and decreased plasma H2O2content(P<0.01),GSH-Px activity and MDA content in broilers(P<0.05).The above results showed that the addition of low dose of Lactobacillus plantarum powder improved the antioxidant function of jejunum to some extent,and the high dose of Lactobacillus plantarum powder improved the intestinal morphology and barrier function of plasma and jejunum of broilers,but had no effect on immune function.It is suggested that the addition amount of Lactobacillus plantarum is 1000 g/t.
