Seaweed polysaccharide is a polysaccharide extracted fromseaweed.At present,a large number of studies have shown that seaweed polysaccharide has biological functions such as antioxidant,antiviral,antitumor,and antibacterial.Related research applications,this research is dedicated to developing the clinical application of seaweed polysaccharides in dogs.The purpose of this experiment was to explore the effects of feeding seaweed polysaccharides on the digestion and absorption of dogs.The experiment selected 18one-year-old healthy beagle dogs and randomly divided theminto control group(basic diet+ starch capsules),low-dose group(basic diet + seaweed polysaccharide capsules 200mg/kg body weight),high-dose group(basic daily Food + seaweed polysaccharide capsules 800 mg/kg);blood samples were collected on the 0th,7th,14 th,21st,and 28 th day of the experiment,and the dogs were anesthetized and sacrificed on the 28 th day to collect the contents of pancreas,duodenum,jejunum,ileum,and cecum.Mainly through the determination of serumbiochemical indicators;serumand pancreatic digestive enzyme activities;the relative expression of EGF(epidermal growth factor)and SS(somatostatin)mRNA and their protein expression levels in the small intestine of dogs;observe the morphological changes of the small intestine,and detect the cecal microorganisms.Changes in microflora to evaluate the effect of seaweed polysaccharides on the digestive function of dogs.The test results are as follows:1.Biochemical tests showed that compared with the control group,the total bilirubin in the low-dose and high-dose groups on the 14 th day was significantly increased.The serumlevels of ALT,AST,TP,ALB,UREA and CREA in each dose group fluctuated within the normal physiological range.2.The detection of digestive enzymes showed that the activities of amylase,lipase and protease in canine pancreas were no different in each group.Compared with the control group,the protease activity of the high-dose group was significantly decreased on the 7th day(P<0.05).3.The morphological observation of the small intestine showed that the small intestinal villi in the control group and the low-dose group were slightly sparsely broken,and the other intestinal segments had little difference in morphology.The small intestinal villi were neatly arranged,with high density and good structural integrity;Compared with the high-dose group,the crypt depth in the duodenal segment was significantly decreased(P<0.05),and the V/C value of the small intestine was increased(P<0.05).There was no difference in the length of the small intestinal villi among the groups.4.The expression of canine EGF mRNA and protein showed that compared with the control group,the relative expression of EGF mRNA in the duodenumof each dose group was significantly increased;the relative expression of EGF mRNA in the jejunumof the low-dose group was decreased.Compared with the control group,the expression of corresponding EGF protein in the duodenum,jejunumand ileumof the low-dose group was significantly increased;the expression of EGF protein in the ileumof the high-dose group was significantly increased.5.The expression of canine SS mRNA and protein showed that compared with the control group,the SS mRNA in the jejunumof the low-dose group was significantly decreased.Compared with the control group,the relative expression of SS protein in the jejunumand ileumof the low-dose group was significantly increased(P<0.05),and the relative expression of the SS protein in the duodenumof the high-dose group was significantly increased.6.Alpha diversity analysis showed that adding seaweed polysaccharide to the diet could significantly improve the diversity,richness and uniformity of canine cecumflora.The proportion of Clostridiumin the cecal contents was significantly decreased(P<0.05),The above results show that seaweed polysaccharide can increase the V/C value of the small intestine,increase the relative expression of EGF mRNA,SS mRNA and EGF protein expression,improve the diversity,richness and uniformity of canine cecal flora,and improve the cecal bacteria.The proportion of beneficial bacteria in the group can improve the digestive function of dogs.
