| Parasitic diseases are one of the most common infections in livestock and pose a greater risk to livestock production.Gastrointestinal nematodes can cause anaemia,diarrhoea and severe nutritional depletion in sheep.In addition,parasitism can have an indirect effect on metabolism and increase susceptibility to other pathogens.Benzimidazole,imidazole and macrolides are widely used to control gastrointestinal nematodes.However,frequent use of the same drugs can lead to the development of drug resistance.This increases gastrointestinal nematode infections,affects sheep performance and reduces the productivity and economic efficiency of the flock.Moxidectin is a highly effective broadspectrum anthelmintic widely used in a variety of animals for the control of gastrointestinal nematodes and surface parasites.Compared to ivermectin,moxidectin has better efficacy,higher distribution,longer elimination half-life and better safety profile.Currently,moxidectin is commonly used clinically in injectable,tablet and transdermal formulations for cattle,sheep,horses and pigs,and is an ideal drug for antiparasitic in vitro and in vivo.In order to achieve the purpose of long-acting and slow release of moxidectin formulation,the group has developed moxidectin gel formulation.In order to study the clinical efficacy and drug safety of Moxidectin gel preparations.The following tests were carried out:1.Clinical efficacy test of moxidectin gel on Qinghai Tibetan sheepThe clinical efficacy trial was conducted in a natural grazing pasture in Gonghe County,Hainan Tibetan Autonomous Prefecture,Qinghai Province,which did not use deworming drugs for 3 months before the trial.Clinical observation found that Tibetan sheep’s diet,drinking water and exercise behavior were not abnormal.From the weaned and adult Tibetan sheep in the pasture,150 Tibetan sheep with similar body weight were selected,with a body weight of 23.63 ± 4.07 kg.The test was randomly divided into 5 groups,30 Tibetan sheep in each group,the moxidectin pouring agent group was poured along the midline of the dorsum at a dose of 0.5 mg/kg;the ivermectin injection group was given at a dose of 0.2 mg/kg The doses were administered by subcutaneous injection;the low,medium and high dose groups of moxidectin gel were administered by subcutaneous injection at doses of 0.2 mg/kg,0.5 mg/kg and 1 mg/kg,respectively.Feces were collected rectally on the 0st and 3rd days after administration,and stored in a refrigerator at 4°C.The fecal egg count method was used to calculate the average number of eggs per gram of feces before and after administration,which was used as the evaluation standard for the clinical efficacy of anthelmintics.The results showed that the average number of eggs per gram of feces in the low,medium and high dose groups of moxidectin pouring agent,ivermectin injection and moxidectin gel before administration were 772.22,833.33 and933.33 respectively.638.24 and 767.65;the average number of eggs per gram of feces after administration were: 178.86,221.22,133.32,0 and 0,respectively.The reduction rates of alimentary canal nematode eggs in each group were 76.84%,73.45%,85.72%,100% and 100%.2.Target animal safety test of moxidectin gel40 weaned healthy Hu sheep were selected,half female,with a body weight of25.73±1.23 kg.Randomly divided into 4 groups(n = 10),namely blank control group,1times dose group(0.2 mg/kg),3 times dose group(0.6 mg/kg),5 times dose group(1mg/kg),given the drug group was given subcutaneous injection for 3 consecutive days,and the feeding and management were carried out according to the normal production management of the farm.On the day 0 and on the 1st,3rd,and 10 th days after administration,the body weight,body temperature,respiration and pulse rate of the sheep were measured,and anticoagulation and non-anticoagulation were collected for blood routine and blood biochemistry Analysis,collection of urine samples to determine urine routine.On the 10 th day of the experiment,according to the principle of half females,4in the control group,4 in the 1-times dose group,6 in the 3-times dose group,and all the experimental sheep in the 5-times dose group were randomly sacrificed,and each sheep was collected.The heart,liver,spleen,lung,kidney,stomach and colon tissues were weighed and the organ index was calculated,and tissue sections of the heart,liver,spleen,lung,kidney,and colon were made,HE stained,and the tissue sections were observed.The results showed that there were no significant differences in other indicators except for a few indicators that were significantly different from the control group within the normal physiological range.Moxidectin gel did not significantly affect the basic indexes,blood routine,blood biochemistry and organ index of sheep,and no abnormal pathological changes were observed in pathological tissue sections.To sum up,moxidectin gel has good deworming effect on digestive tract nematodes in Qinghai Tibetan sheep at low,medium and high doses.In the case of 1,3 and 5 times the recommended dose for three consecutive days,the sheep did not have adverse reactions,and the body weight,blood routine,blood biochemistry and other indicators were also within the normal range,and no abnormality was found in tissue sections pathological changes. |
