Edible fungi are favored by researchers and cultivators because of their strong ability to convert exogenous inorganic selenium accumulation into bioavailable and easily absorbed selenium.Mushroom is one of the common edible fungi species cultivated in China,with high nutritional and medicinal value.Its main active ingredient,lentinan,has antioxidant,anti-tumor and immunity-enhancing activities."Selenium" is one of the essential trace elements in the human body,and it is an important part of a variety of important selenium-containing enzymes in the human body.In this study,four kinds of mushrooms,including Shenxiang "215",Shenxiang "1504",Shenxiang "818" and "Xiangyang No.2",were used to conduct experiments on mushroom mycelium and original seed cultivation stage to understand the enrichment of selenium in mushrooms,and at the end the varieties with strong selenium-enriching ability are selected and the optimal concentration ratio of exogenous selenium addition is determined.The research results have certain theoretical and technical support significance for understanding the physiological characteristics of selenium-enriched mushrooms,understanding the characteristics of selenium-enriching cultivation,and guiding selenium-enriching cultivation techniques.practical value.The results showed that:1.Characteristic analysis of selenium collection during the culture period of solid strain mycelium For solid mushroom strains,according to the observation of adding different selenium agents,under the condition of temperature 23~27℃,the growth rate of mycelium with yeast selenium is faster,and the optimal concentration is 60~75mg/kg(60mg~75mg/L),showing dense white hyphae,high density,and complete colony morphology.Under the same conditions,the growth rate and quality of the samples added with yeast selenium were generally better than those added with sodium selenite(Na2SeO3).There are differences in the tolerance of different solid varieties to selenium,for example:Shenxiang 215 has poor tolerance to selenium on the third day,and the amount of 75%selenium in yeast on the seventh day is faster than that in the control group.The overall phenomenon is to inhibit first and then promote growth.In general,Shenxiang 1504 showed a faster growth rate than the control group after adding selenium,and the growth rate of yeast selenium 60%was the fastest.The Shenxiang 818 strain showed that the growth rate of mycelium after adding selenium was faster than that of the control group,and the average growth rate was the fastest with the addition of 75%yeast selenium.The overall mycelium growth of Xiangyang 2 was strong,and the average growth rate of the sample group added with 60%yeast selenium was the fastest.After the domestication of the parent species,the optimum amount of yeast and selenium was inoculated,and a total of 1500 rods were produced.The key technical points of cultivation temperature,humidity,light,and pest control were summarized.According to the fruiting yield and high-quality shiitake mushrooms,according to the yield analysis,the yields of the four kinds of mushrooms are ranked as follows:Xiangyang No.2>Shenxiang 215>Shenxiang 1504>818.The yield of Xiangyang No.2 is the highest,reaching an average of 1.84 kg per stick,and the mushrooms are thick and fleshy.According to the results of the detection of selenium content,the varieties with higher selenium content were screened out:Shenxiang 818>Shenxiang 215>Xiangyang No.2>Shenxiang 1504,the highest selenium content detected in the fruit body reached 0.64 mg/kg.2.Characteristic analysis of selenium collection in liquid medium myceliumKeeping other conditions the same,the yeast selenium at the same depth in the CPDA medium,the four strains(Shenxiang 215,Shenxiang 1504,Shenxiang 818,and Xiangyang 2)were in the same selenium-enriched medium with a concentration of 60 mg/kg.And the culture environment and growth time are exactly the same.After the mycelium is full of the petri dish,it is moved to the liquid medium for cultivation,the mycelium is collected,dried,and the biomass is measured to determine the total selenium content.According to the test results,Shenxiang 215 has the strongest ability to collect selenium.At the concentration of 60mg/kg,the biotransformation of Shenxiang 215 was 10.27mg/kg.The seleniumcollecting ability of liquid strains is far greater than that of solid strains.
