Twig compost composting agent is a composite microbial agent composed of a variety of microorganisms with lignin and cellulose degrading functions after high temperature acclimation.It has research value in promoting composting rate and improving compost quality.The research is mainly based on the research object of the compost composting agent of twigs.The fermentation process of composting agent,the degradation ability of lignocellulose and the application effect of compost are analyzed and researched respectively.In the research of fermentation technology,a combination of single factor experiment and orthogonal experiment is used to determine the effective viable count,laccase(Lac),lignin peroxidase(Lip),and manganese peroxidase(Mnp).Enzyme activity is the screening index,the fermentation process of the composting agent is optimized;the treatments with different inoculum amounts are set,and the 70 d composting agent is used to degrade lignocellulose;the stripe-coated aerobic composting mode is used to set up the experimental composting agent,Commercially available composting agent and blank experiment three groups of treatments,measuring temperature,water content,p H,EC,nutrients,germination index,lignocellulase activity and other physical and biological indexes during the composting process,and analyzing the influence of composting agent on the composting process.The results of the study are as follows:1.The optimum medium composition for branch compost composting agent is 80% bran,10% biogas residue,10% corn meal,0.25% glucose,0.25% tryptone,0.05% magnesium sulfate,0.01% calcium chloride;optimal fermentation The conditions are that the solid-liquid ratio is 1:1,the inoculum is 25%,and the temperature is 30℃;after the fermentation process is optimized,the effective number of viable bacteria of the decomposing agent can reach 7.50×1010cfu/g;Finally,a powdery decomposing agent that meets the technical specifications and standards can be obtained.2.The functional enzyme activity in the fermentation process is mainly and effective The number of viable bacteria is related,the more effective viable cell count,the stronger the enzyme activity;Mg2+ and Ca2+have a certain effect on the enzyme capacity of lignin degradation.The addition of Mg2+ can induce more lignin peroxidase,the enzyme activity can reach 20.50U/g,and the addition of Ca2+ can induce more lacquer Enzyme,the enzyme activity can reach 16.88U/g;3.The inoculation amount of branch compost composting agent has the best degradation effect at 0.2-0.25%,the highest cellulose degradation rate is 57.45%,the highest degradation rate of lignin is 53.18%,and the degradation is completed in about 20 days;They were 2.93 and 2.48 times that of CK treatment,and 80% of the total degradation was completed in about 20 days.4.The treatment with branch compost composting agent was added to the high temperature stage 1day earlier than the commercial composting agent and the CK treatment.The high temperature stage lasted for26 days,which was 3 days longer than the CK treatment.After 30 days of composting,the water content was 38.16%,and the germination index was 119.93%,and its moisture content changes with the added commercial composting germination index changes similar agents,better than CK treatment;which P and K as compared to other treatments significantly increased,increased by 0.60% and 1.18%.5.The treatment of adding branch compost composting agent has a strong carbon source utilization capacity in the entire composting process.Except for the initial stage,the AWCD value of each stage is significantly higher than other treatments;Shannon index is higher than other treatments,and Simpson index is basically stable;in PCA analysis,compost decomposition treatment agent is added shoots the main component with other treatment coordinate system differences are significant,the various stages of composting a main component in the coordinate system distributed more concentrated.It shows that the functional bacteria in the compost composting agent can play a continuous,stable and efficient role in the composting process.In summary,the twig compost composting agent obtained by fermentation process optimization has a high effective number of viable bacteria,has excellent lignin and cellulose degradation functions,and shows fast heating,high activity,stable action,and high temperature adaptability in composting applications.And the characteristics of strong degradation ability,high quality compost product and high phosphorus and potassium content.Compared with common compost composting agents,it has more advantages in the application of branch composting.
