Benzo(a)pyrene(BaP)is a widespread and harmful substance that can affect the reproductive performance of female mammals in many aspects.However,it is not clear whether BaP affects the maturation and subsequent development potential of oocytes in livestock.In this study,the effects of BaP on the maturation and developmental potential of oocytes were studied in mice.On this basis,the recovery of potential damage of oocytes induced by BaP by luohangoglycoside V(MV)was further investigated.The results are as follows:Female mice were exposed to BaP by gavage and results showed that BaP resulted in increased rates of abnormal parental oocytes and zygotes,decreased rates of two-cell and blastocyst development,and a decreased number of offspring.Although BaP did not affect the modification of H3K4me2,H3K9me2,H3K27me2,5mC,and m6A in oocytes,BaP led to the modification of H3K9me2,H3K9me2,H3K27me2,5mC,and m6A in zygotes and the abnormal expression of corresponding genes.In addition,female mice exposed to BaP significantly reduced the maturation rate of oocytes in offspring,and increased the rates of spindle abnormality and chromosome abnormality.Meanwhile,the content of mitochondria and ATP in oocytes decreased,the early apoptosis rate,oxidative stress level and 5mC level increased,and the embryo development rate significantly decreased.Mice were given both BaP and MV,and the results showed that MV could reduce the effects of BaP on oocyte maturation,spindle morphology,chromosome arrangement,acetylated tubulin,actin,Juno protein levels,reactive oxygen species levels,and repair DNA damage and reduce apoptosis.Furthermore,MV alleviates the inhibition of BaP on early embryonic development.In summary,the results of this study revealed that BaP caused abnormal maturation and decreased developmental potential of parental and offspring oocytes in mice.MV can protect the damage of BaP on oocytes.The results of this study will help us to better understand the effects of pollutants on the breeding performance of livestock and provide reference for the avoidance of harmful substances in livestock production.
