As a kind of herbaceous plant originated in China,the seed kernel of Coix is a kind of traditional Chinese medicine and small miscellaneous grain,and the whole plant has excellent feeding value.Coix seed shell is located in the outermost layer of seed,developed from abnormal leaf sheath,the scientific name is involucre,its function is similar to corn bract leaf,it has a very good protective effect on seed kernel.The type of seed shell is the biggest difference between wild Coix and cultivated Coix.The shell of wild Coix is stone,thick and hard,and the cultivated species are paper,thin and fragile.The results of previous studies in this laboratory showed that the stone shell of Coix lacquer was controlled by a single dominant gene.It was previously named the physical dormant gene Ca Sdr1 based on the function of the gene.n this study,the genetic linkage map of Coix was constructed by using cultivated Coix(XBK)and wild domesticated variety(DHS)as parents,and the physical dormant gene Ca Sdr1 was located in the 383 Kb region.The main results are as follows:1.Using 551 F2 plants and 248 In Del markers,the genetic map of Coix was constructed,which was divided into 10 genetic linkage groups with a total length of 1682.73 c M,and the average genetic distance between markers was 6.79 c M.2.The collinearity between the genetic linkage map and the physical map of Coix lacquer showed that most of the marker loci were colinear with the genome sequence of the physical map.Of the 248 marker loci in the 10 linkage groups,230 were in accordance with the physical order of the genome of DHS,covering 86.81% of DHS genome.3.In the initial mapping interval,a new polymorphic marker was developed.Using 3Indel markers and 1 SNP marker,793 exchange markers were screened out from 16113 F2 plants.The physical dormant gene of Coix lacquer was mapped between marker Indel10893-PK1 based on the phenotypic 113 exchange single-plant and two-wing marker genotypes with a physical distance of 383 Kb.
